The paper presented the application advance of mass spectrometry techniques on Chinese medicine research from three aspects: the component analysis, bio-activity screening and metabonomics research. The section of the component analysis focused on various methods for analyzing oligosaccharide isomers. And the applications of the fingerprint technique of mass spectrometry to the component analysis and quality control of Chinese medicine were also provided. On the research of bioactivity compound screening, some techniques were introduced respectively, including the ultrafiltration-mass spectrometry, cell membrane chromatography-mass spectrometry, microdialysis probe-mass spectrometry, affinity chromatography-mass spectrometry, intensity-fading mass spectrometry, modified agarose-mass spectrometry and direct analysis mass spectrometry, etc. On the metabonomics studies, we provided some examples which clarified the action mechanism of Chinese medicine on hepatotoxicity, kidney deficiency, myocardial infarction and type 2 diabetes. The above content reflected the importance of mass spectrometric techniques in the innovative research of Chinese medicine.

To study the content variation of ginsenosides after the combination of ginseng with Zingiber officinale Rosc or Radix Paeoniae Rubra,a high performance liquid chromatography-electrospray ionization spectrometric(HPLC-ESI-MS) method was used to analyze and compare the ginsenosides in the decoction. The experimental results show that the contents of ginsenosides decreased along with the increased amount of Zingiber officinale Rosc, while the stripping of ginsenosides was promoted by a small amount of Radix Paeoniae Rubra. In addition, the total antioxidant activity of n-butanol and water extracts from the single and compatible decoctions of ginseng was also determined by ferric reducing antioxidant power(FRAP) method. With ascorbic acid(500 μmol/L) as control, the anti-oxidation of compatible decoction of ginseng with Zingiber officinale Rosc/Radix Paeoniae Rubra was higher than that of the single decoction of ginseng;and the FRAP values of n-butanol extracts from the compatible decoction of ginseng with Zingiber officinale Rosc or Radix Paeoniae Rubra were 1562.29 and 2969.78 μmol/L,respectively,greater than the sum of single decoction of ginseng with Zingiber officinale Rosc(1260.27 μmol/L) /Radix Paeoniae Rubra(2502.07 μmol/L),indicating that the antioxidant activity of n-butanol extract from compatible decoction was better.

Abstract The applications of mass spectrometry (MS) and its coupling technologies (gas chromatography-MS, high-performance liquid chromatography-MS, capillary electrophoresis-MS, etc.) on the chemical constituents of lignans, essential oils, organic acids, and triterpenoids in ( Schisandra chinensis and Schisandra sphenanthera ) fruits are reviewed and their potential applications are summarized. The aim of this study is to provide a basis for rapid screening of chemical constituents from Chinese medicine, identification of medicinal material varieties, and quality control methods.

Inhibiting effect of the curcumin(Cur) in combination with cis-diamminedichloroplatinum(DDP) on proliferation of COC1 cells was analyzed with MTT colorimetric assay. The morphological changes of COC1 cells were observed by inverted microscope. The result indicated that DDP of 2.5~10 mg/L dose combined with curcumin(Cur) of 5 mg/L dose could enhance the inhibiting effect and induce apoptosis compared with the case when only DDP was used.Meanwhile, curcumin of 5 mg/L dose could improve the sensitization of COC1 cells to DDP with 2.66 times.Optical microscopy results showed that COC1 cells had a marked decreased proliferation and a great amount of cells necrosis in DDP+Cur group and DDP group than the control group.COC1 cells had lower diopter, scatter array, serious vacuolization treated with DDP+Cur.The HPLC result indicated that the contents of DDP within tumor cells decreased lightly after dosage of Cur,but it had no statistically significant(P0.05).In summary, curcumin combine with DDP can enhance the inhibiting effect on proliferation of COC1 cells, meanwhile, curcumin can improve the sensitization of COC1 cells to chemotherapeutics. However, the curcumin is unlikely to affect the amount of DDP within cells.
