Urinary estrogens I improved method for quantitation of urinary estrogens

Abstract A specific method for quantitation of the three classical urinary estrogens, estrone, estradiol-17β, and estriol is described. The method involves conjugate extraction, enzyme hydrolysis, phenolic separation, thin layer chromatography of the phenolic steroids in a 20% acetone-80% isopropylether system and finally separation and quantitation by gas chromatography. A formation of derivatives is not required. The precision, sensitivity, and specificity of the method are evaluated. Examples of the application of this method are evaluated. Examples of the application of this method are described.

Chromatographic properties of the epimeric estriols

Abstract Thin-layer and gas chromatographic methods to separate the epimeric estriols are described. Gas chromatography of the acetates provided separation of all four. Thin-layer chromatography on Silica Gel G separated the cis from the trans estriols. The trans epimers separated only on silica gel modified with ammonium bisulfate. The acetonides separated well on both thin-layer and gas chromatography.

Urinary estrogens II excretion of estrogens in rhesus monkeys stimulated with human gonadotropins

Abstract Spontaneously non-cycling rhesus monkeys were treated with gonadotropins extracted from human pituitaries and an HMG preparation. Treatment was carried out for 10 – 11 days with 75 IU FSH daily followed by a single injection of 2000 IU LH. Daily urinary estrogen levels were determined by a method involving conjugate extraction, enzyme hydrolysis, and phenolic separation followed by two thin layer separations and gas chromatography. Estrone was found to be the major urinary estrogen. Estradiol-17β and estriol were detectable only in small quantities coinciding with high concentrations of estrone. Estrone levels as high as 56 μg/24 hours were found when multiple ovulations occurred. Ovulation time and ovarian size were determined by laparotomy.

A new system for paper chromatography of polar steroids

Abstract The new paper chromatographic system, isopropyl ether-ethylene glycol system, provides an improved and faster separation of steroids, particularly estrogens in paper chromatograms. The mobilities of a number of steroids are listed.
