Factors affecting repair of sublethal injury in frozen or freeze-dried bacteria

Abstract Conditions facilitating recovery of sublethally injured cells present in frozen or freeze-dried preparations of bacteria were investigated. Some cells of Streptococcus faecalis injured by freezing required RNA resynthesis for recovery, whereas the rest did not. Addition of 6% NaCl to control recovery medium was found to inhibit RNA synthesis completely in frozen-injured cells of S. faecalis, but not in intact ones. This seems to be a principal reason why 6% NaCl inhibits the growth of the injured cells. The recovery of viable cells of freeze-dried and stored Streptococcus thermophilus was greately increased by the addition of cysteine to the plating medium. The favorable effect of cystein was mainly due to its reducing activity. On the contrary, the addition to the plating medium of certain types of peptone, such as lactalbumin hydrolyzate, resulted in a considerable decrease of viability in frozen preparations of Vibrio metschnikovii. Thus, at present, it seems too early to try to discuss specifically the optimal composition of plating media to give the highest viability from frozen or freeze-dried preparations of bacteria.

Usefulness of nutrient agar supplemented with NaCl in detection of heat-injured Escherichia coli.

We examined the effect of composition of agar medium for the detection of viable cells of Escherichia coli JCM1649T. The nutrient agar supplemented with NaCl (NA) and two commercial products of plate count agar (standard method agar) exhibited practically the same level of colony count for intact cells, but NA was the most suitable for the detection of sublethally injured cells induced by heat treatment at 55°C for 15 min. Addition of 0.5% NaCl to the commercially-available plate count agar improved the detection rate of heat-injured cells, while glucose in the agar medium was found to decrease the detection rate. NA was shown to be useful as repair medium in the selective detection of heat-injured E. coli.

Effect of Temperature, Moisture and Fat Contents on the Thermal Conductivity of Meats.

熱伝導率の測定法には定常法と非定常法がある,挽肉(牛肉,豚肉,鶏肉)に脂肪を配合して平行熱源法と線熱源法を用いて5~60℃の温度域における熱伝導率を測定した.その結果,平行熱源法では温度上昇に伴って熱伝導率がわずかに上昇する傾向が観察されるのに対し,線熱源法では温度上昇による影響が少ないことが認められた.また,水分含量と熱伝導率との間には正の指数比例関係,脂肪含量と熱伝導率との間には負の指数比例関係があり,この2成分因子から熱伝導率を求める推定式は牛肉では次のとおりであった.ただし,λp, λhはそれぞれ平行熱源法及び線熱源法による熱伝導率(W/(m・K)),x1…水分(%),x2…粗脂肪(%).

Limit of Application of Breed Method and Relationship between Individual Bacterial Counts measured by Breed Method and Colony Counts measured by Standard Plate Count Method

生乳中の細菌数の測定で, Breed法の適用性の限界, 並びにBreed法による直接鏡検個体数 (全菌数) と標準平板培養法による生菌数との関係を, 別報のBactoscan 17600 (A/S N. Foss Electric, Denmark) の性能試験で得た成績を用いて検討した.生乳305点で, Breed法で求めた全菌数の対数値とBactoscanのインパルスのそれとの相関係数は, 0.936であったが, これをBactoscanのインパルス1000 (約50万/mlの細菌数に相当) 以下の場合, 1000~10000の場合, 10000以上の場合に分けると, それぞれ, 0.491 (n=70), 0.809 (n=144), 0.824 (n=91) となり, 最初のものは, 著しく低い値を示し, Breed法の適用性の限界が示唆された. また, Bactoscanのインパルスで1000以上を与える試料について, 全菌数の対数値と生菌数のそれとの相関係数は0.819であった. 生菌数を何倍すれば, 全菌数に見合う数値になるか調べるため, 生菌数に種々のファクターを乗じた値を求め, 対応する全菌数の, この積に対する比率の度数分布を比較した. この結果, ファクターが3.5の場合, 全菌数/(生菌数×ファクター) で計算される比が0.33~3.0の範囲に入る試料の (点数の) 割合が84.7%と最大となり, かつヒストグラムの形も左右対称に最も近かった. 即ち, 生菌数に, 3.5を乗じた値で, 直接鏡検個体数のオーダーを把握できることが明らかとなった. そして, この値は生乳中に分布する細菌の菌塊 (コロニーの形成単位) の平均個体数に相当すると考えられる.

TTC test on individual milk samples

個乳にTTCテストを適用した場合の問題点を検討するため,抗生物質を与えていない34頭のホルスタイン牛の個乳試料181点についてTTCテストを行なったが,TTCの還元が認められず抗生物質陽性と判定されるものが3点あった.これらはいずれも乾乳直前の末期乳であった.しかし末期乳でも異常の認められない例の方が多かった.したがって末期乳には抗生物質の移行残留がないにもかかわらずTTCテスト陽性の反応を示すものがある点に留意すれば,個乳に対してTTCテストを適用しても問題はない.一部の末期乳がTTCテスト陽性の反応を呈するのは,検定菌Str. thermophilus 510の発育をわずかに阻害する物質がホエー部分に含まれていることによると考えられる.

Nutritional Requirements of Lactobacillus bulgaricus

Cultures of Lactobacillus bulgaricus often fail to grow on media commonly used for lactic acid bacteria such as standard plate count agar. This is considered dus to deficiencies of some growth factors required by the organisms. To determine the factors deficient in these media, a survay of growth requirements of six cultures of L. bulgaricus in chemically defined media was done.All of six cultures required “tween 80” and enzymatic digest of casein for their growth. “Tween 80” could be replaced with “tween 40” plus oleic acid, lecithin or cephalin.Three cultures required pantethine in a medium containing 0.4γ/ml of calcium pantothenate. However, pantethine could be replaced with higher amounts (e.g., 1γ/ml) or calcium pantothenate. When cysteine level in a medium was reduced to 10γ/ml (one-tenth of usual synthetic media) and cystine was omitted, calcium pantothenate in a concentration of 2γ/ml failed to replace pantotheine. This suggests that “activity coefficient” pantethine/pantothenate is a function of cysteine level in a medium.One culture identified asThermobacterium jugurtOLRA-JENSEN did not grow in the absence of lactose even after prolonged incubation period.None required erotic acid or thymidine, and the latter seemed to be inhibitory to some cultures.

Aroma Production of Lyophilized Lactic Cultures

Str. cremorisとLeuc. citrovorumの混合したバタスターターおよびStr. diacetilactisの牛乳培養を凍結乾燥して30°Cに保存し,凍結乾燥菌の芳香成分(biacetyl+acetylmethylcarbinol)の生産性について実験した. 保存期間が長くなるに従って生菌数の減少および生酸活性の低下によって芳香成分の生成は遅延するが,凍結乾燥菌を牛乳に培養した場合,酸度と芳香成分生成の関係はもとの培養と比較して特に差は認められなかった.また40~50日保存した凍結乾燥菌の牛乳培養における芳香成分の生産量ももとの培養と差はなく,乳酸菌の芳香生産性は凍結乾燥によって全く影響をうけないことが確認された.

(1) 酪農乳酸菌Streptococcus cremoris H-61およびLactobacillus bulgaricus B-1の脱脂乳培養を凍結乾燥して,その生残率と乾燥菌保存中の生菌数の変化を検討した. (2) 凍結乾燥における生残率は,菌株や培養の状態によって相違し,培養を脱脂乳で希釈すると生残率が高まりStr. cremoris H-61では60%, L. bulgaricus B-1では40%前後の生残率を示した. (3) 凍結乾燥乳酸菌は保存中に対数的に死滅するとみなすことができ,その死滅率で乾燥菌の保存性を定量的に比較できた. (4) 培養に脱脂乳を加えて希釈すると希釈度の高いほど,保存中の死滅率は減少した.また培養のpHを調整すると死滅率が変化し, Str. cremoris H-61ではpH 6~9, L. bulgaricus B-1ではpH6附近で最小の値を示した. (5) 乾燥乳酸菌の死滅率の見かけのQ10は2.2~3.5(10~37°)であった.
