Two-dimensional polyacrylamide gel electrophoresis of bovine seminal plasma proteins and their relation with semen freezability

The objective of this study was to evaluate the low weight (10-30 kDa) protein profile of bovine seminal plasma using two-dimensional polyacrylamide gel electrophoresis (2D-PAGE) and to determine if any of these proteins was associated with semen freezability. Seminal plasma was collected from 16 bulls of high or low semen freezability. Twelve protein spots were identified from the 2D gel (15%); six of these were present in all samples. Of the 12 proteins found, three spots, present in all samples, 3 (15-16 kDa), 5 (16-17 kDa), and 7 (10-12 kDa) had nonsignificant variation among bulls, regardless of their freezability classification. Four proteins were more abundant (P<0.05) in seminal plasma samples collected from bulls with high semen freezability than in samples of bulls with low semen freezability: the spots 3 (15-16 kDa, pI 4.7-5.2), 7 (11-12 kDa, pI 4.8-4.9), 11 (13-14 kDa, pI 4.0-4.5), and 23 (20-22 kDa, pI 4.8-5.2). On the other hand, spot 25 (25-26 kDa, pI 6.0-6.5) was more abundant (P<0.05) on seminal plasma samples from bulls with low semen freezability. The N-terminus sequence of protein 7 was identical to the acidic seminal fluid protein (aSFP). Protein 23 (after trypsin digestion) had structural similarity to bovine clusterin. We concluded that there were differences in the seminal plasma protein profile from bulls with low and high semen freezability; aSFP, clusterin, proteins 3 and 11 may be used as semen freezability markers; and protein 25 was related to low semen freezability.

Relação entre a condição corporal e a idade das ovelhas no encarneiramento com a prenhez

In this study, the relationship between score of body condition (BC) at the beginning of mating and ewes age with the pregnancy rate (PR) of a Corriedale sheep flock, grazed in the State of Rio Grande do Sul- Brazil are presented and discussed. The BC and PR observed on the flock were 90.4% and 2.84(±0.57) respectively. The mean of BC of the pregnant (P) and non-pregnant (NP) ewes was P 2.86 (±0.56) and NP 2.64(±0.59). The values of P and NP have shown statistic significance (P< 0.001). The results showed, also, that the PR increase as the BC increases, reaching 92 and 98% on the group of ewes with BC 3.0 and 4.0 respectively. Finally, the regression logistic analysis from the data showed that the age of the ewes have no relation with the PR and a positive relationship between pregnancy and body score condition ( p=0.002).

IgE sérica total em atópicos e não-atópicos na cidade de Porto Alegre

PURPOSE: We determined the total serum IgE levels among healthy individuals and atopic patients with respiratory symptoms in Porto Alegre. METHODS: Atopic patients (n: 92) and normal controls (n: 86) were stratified according to age and sex. Presence of atopy was recorded by history, clinical examination and skin prick test reactivity to common aeroallergens. Serum IgE was measured by Pharmacia Immuno-Cap System. RESULTS: It was demonstrated that total serum IgE is higher in males than in females. Total serum IgE levels were significantly higher in the atopic group (404 UI/mL) in comparison with the non-atopics controls (54.4 UI/mL). Our study provided the normal range for total serum IgE among atopics and non-atopics individuals in Porto Alegre. CONCLUSIONS: The profile of serum IgE among our population (atopics and non-atopics) is similar to the one observed in other countries.

Prevalence of antibodies against chicken anaemia virus (CAV) in broiler breeders in Southern Brazil

Chicks infected during the first two weeks of life with chicken anaemia virus (CAV) manifest clinical disease that can be avoided if the breeder hens transfer enough antibodies to their progeny. The objective of the present work was to establish the prevalence and titer of anti-CAV antibodies in some Brazilian broiler hen breeder flocks and verify in which phase of life the birds were infected. A total of 1,709 serum samples from 12 broiler hen flocks vaccinated against CAV and 64 unvaccinated flocks were analyzed for CAV antibodies with an enzyme-linked immunosorbent assay (ELISA). All non-vaccinated breeder flocks were found to be infected with CAV, with 89% of the hens tested presenting antibodies, 52% of these with titers considered high enough to protect their progeny against CAV infection. Likewise, all vaccinated hens had antibody titer to CAV capable of conferring protection to their progeny. Thus, vaccination of hens seems capable of conferring protection to chicks against clinically apparent CAV-associated disease.

Análise da contaminação por Salmonella em ovos do tipo colonial através da reação em cadeia da polimerase

The identification of salmonella infection in commercial poultry has been one of the strong points of prophylaxis and consequent reduction of salmonellosis outbreaks in humans associated to consumption of eggs, considering that the analysis of the eggs can be one more point of detection of infection, which for many times appear without clinical signs. The Polymerase Chain Reaction (PCR) seems to be a useful strategy for Salmonella detection, because various authors have used the PCR to verify the presence of bacteria in meat, feces, tissues, blood, milk and eggs, with different methods of manipulation of samples. We have analyzed 360 eggs from ten farms, producers of free range-eggs, in the district of Camobi, in Santa Maria n RS n Brasil. The eggs were grouped in pools of six, totaling sixty samples. The bacteriological exam was done in compliance with the method preconized by the technical rules and the method for extraction of DNA was by phenol-chloroform. The PCR was performed for the amplification of a 284 bp DNA fragment. The analysis of the results do not show significant difference between the PCR and the bacteriological exam. Feeding dairy cows with soybean by-products: effects on metabolic profile

Soybean by-products are currently used in Rio Grande do Sul, Brazil, as a source of protein in dairy cows. However, the high protein breakdown of this feed source in the rumen causes loss of the intrinsic quality of the protein and increases plasma urea with deleterious consequences to animal metabolism. Thermal treatment of soybean can overcome this limitation. The aim of this study was to evaluate the metabolism of dairy cows fed with thermally treated soybean and raw soybean, through the analyses of the metabolic profile of plasma and milk. Twelve Holstein cows in mid-lactation period were studied using four treatments: commercial concentrated of protein, soybean meal, raw soybean and roasted soybean. Protein and urea levels in milk were measured in morning and afternoon samples. Data were arranged in a Latin square design (4 treatments and 3 animals in each square). There were no significant differences in glucose, beta-hydroxybutyrate, calcium, phosphorus and magnesium concentrations among treatments. Cows consuming raw and thermally treated soybean had higher levels of plasma cholesterol. Cows consuming roasted soybean had lower level of plasma and milk urea in the morning. Afternoon milk samples had higher levels of urea than morning samples. It is suggested that thermally treated soybean was effective in diminishing the breakdown of protein in the rumen. It is better to employ morning milk samples to evaluate metabolic profile than afternoon samples.

Determinação do volume saturante de solução desinfetante em piso de chão batido de granja avícola em função da carga microbiana (coliformes) e composição do solo

The floor soil ground is used in most of the Brazilian poultry farms due to the high cost of the construction of concreted floors. The presence of organic matter on the surface of the soil hinders its disinfection, due to the reduction of the antimicrobial action of disinfectants and the tests accomplished in vitro to verify the effectiveness of the disinfectants do not contemplate the adverse conditions found at poultry farms. Thus, the present work intended to determinate the volume of disinfectant solution required to be used on m 2 floor and the counting of total and fecal coliforms in floor soil ground. Presence of total and faecal coliforms was detected in the depth of 0.5cm with 1100NMP g -1 e 460NMP g-1, respective. The volume of disinfectant solution defined to saturate 1m² to 1cm of depth was of one liter.

Performance comparison between broilers positive and negative for antibodies against the chicken anemia virus

The chicken anemia virus (CAV) is present in virtually every country investigated, Brazil including. The aim of this study was to determine what the difference in performance is between positive (progeny of breeders vaccinated or with natural infection) and negative broilers to the presence of antibodies against the CAV in currently intensive raising systems. As a result, it was observed that negative broilers were significantly heavier than positive broilers. Negative males had a final weight 5.43% higher than positive males. There was no significant difference among different treatments in relation to parameters as mortality and feeding conversion. These study indicated that the presence of antibodies against CAV in broilers – may it be through vaccination or natural infection of breeders – did not generate progeny with superior performance under the tested raising conditions.
