Colonized Aedes albopictus and its sexual performance in the wild: implications for SIT technology and containment

BackgroundMating is a physiological process of crucial importance underlying the size and maintenance of mosquito populations. In sterile and incompatible insect technologies (SIT and IIT), mating is essential for mass production, persistence, and success of released individuals, and is a central parameter for judging the effectiveness of SIT/IIT programs. Some mosquitoes have an enormous reproductive potential for both themselves and pathogens and mating may contribute to persistence of infection in nature. As Aedes albopictus can transmit flaviviruses both sexually and horizontally, and as infected insects are usually derived from laboratory colonies, we investigated the implications of mating between a long-term laboratory colony of Ae. albopictus and wild populations.MethodsThrough a series of mating experiments, we examined the reproductive outcomes of sexual cross-affinity between laboratory-raised and wild adults of Ae. albopictus.ResultsThe results indicated appreciable mating compatibility between laboratory-reared and wild adults, and equivalent levels of egg production among reciprocal crosses. We also observed comparable larval eclosion in lab females mated with wild males, and increased adult longevity in female offspring from wild females|×|laboratory males crosses.ConclusionsTaken together, these data suggest that Ae. albopictus can preserve its reproductive fitness over a long period of time in the laboratory environment and has valuable attributes for SIT application. These observations together with the ability to successfully inseminate heterospecific females indicate the potential of Ae. albopictus to act as an ecological barrier if non-sterilized males are massively released in areas occupied by Aedes aegypti. The observed substantial reproductive fitness combined with the capability to reproduce both, itself and viruses illustrates the potential of Ae. albopictus to pose a serious threat if infected and released accidentally.

Household survey of container-breeding mosquitoes and climatic factors influencing the prevalence of Aedes aegypti (Diptera: Culicidae) in Makkah City, Saudi Arabia

OBJECTIVE To investigate the prevalence of container breeding mosquitoes with emphasis on the seasonality and larval habitats of Aedes aegypti (Ae. aegypti) in Makkah City, adjoining an environmental monitoring and dengue incidence. METHODS Monthly visits were performed between April 2008 and March 2009 to randomly selected houses. During each visit, mosquito larvae were collected from indoors and outdoors containers by either dipping or pipetting. Mosquitoes were morphologically identified. Data on temperature, relative humidity, rain/precipitations during the survey period was retrieved from governmental sources and analyzed. RESULTS The city was warmer in dry season (DS) than wet season (WS). No rain occurred at all during DS and even precipitations did fall, wetting events were much greater during WS. Larval survey revealed the co-breeding of Aedes, Culex and Anopheles in a variety of artificial containers in and around homes. 32 109 larvae representing 1st , 2nd, 3rd, and 4th stages were collected from 22 618 container habitats. Culicines was far the commonest and Aedes genus was as numerous as the Culex population. Ae. aegypti larval abundance exhibited marked temporal variations, overall, being usually more abundant during WS. Ten types of artificial containers were found with developing larvae. 70% of these habitats were located indoors. 71.42% of indoor containers were permanent and 28.58% was semi-permanent during WS. Cement tanks was the only container type permanent during DS. Ae. aegypti larval indices (CI, HI, BI) recorded were greater during WS. CONCLUSIONS Taken together, these results indicate a high risk of dengue transmission in the holy city.

Can Adults of the New Zealand Mosquito Culex pervigilans (Bergorth) Detect the Presence of a Key Predator in Larval Habitats

ABSTRACT: The influence of predators on mosquito populations may be direct through predation or indirect through sub-lethal responses of adult mosquitoes in life history traits such as oviposition behavior. In New Zealand, the backswimmer, Anisops wakefieldi, is a common predator of mosquito larvae found in temporary and permanent water bodies. We predicted that the New Zealand native mosquito, Culex pervigilans, whose larvae are vulnerable to predation of Anisops, would likely avoid the containers with the presence of Anisops or its kairomone. We established temporary water containers without predators, free-roaming predators, caged predators (which were unable to eat mosquitoes), or containers from which predators were removed immediately prior to the experiment (these containers would have remnant kairomones from the predators). Each treatment with Anisops had predator densities of one, three, or nine Anisops. Contrary to our predictions, when choosing oviposition habitats, Cx. pervigilans appeared to ignore the presence of free-roaming Anisops, caged Anisops, and water with Anisops kairomone. We thus observed no significant differences between the numbers of egg rafts laid by Cx. pervigilans in the different predator treatments nor were the number of egg rafts significantly affected by the density of predators. Rather than the presence of predators, environmental factors including temperature, humidity, and pressure were significantly correlated with mosquito oviposition. These mosquitoes appeared to either ignore the presence of the predator, had no ability to detect the presence of predators, or perhaps the cues from Anisops predators were not sufficiently strong enough to alarm these mosquitoes. We argue that the mosquito has not evolved the ability to detect the presence of these predators while ovipositing.

Indirect effects of cigarette butt waste on the dengue vector Aedes aegypti (Diptera: Culicidae).

Despite major insecticide-based vector control programs, dengue continues to be a major threat to public health in urban areas. The reasons for this failure include the emergence of insecticide resistance and the narrowing of the spectrum of efficient products. Cigarette butts (CBs), the most commonly discarded piece of waste, also represent a major health hazard to human and animal life. CBs are impregnated with thousands of chemical compounds, many of which are highly toxic and none of which has history of resistance in mosquitoes. This study was performed to examine whether exposure to CB alters various biological parameters of parents and their progeny. We examined whether the mosquito changes its ovipositional behaviors, egg hatching, reproductive capacity, longevity and fecundity in response to CB exposure at three different concentrations. Females tended to prefer microcosms containing CBs for egg deposition than those with water only. There were equivalent rates of eclosion success among larvae from eggs that matured in CB and water environments. We also observed decreased life span among adults that survived CB exposure. Extracts of CB waste have detrimental effects on the fecundity and longevity of its offspring, while being attractive to its gravid females. These results altogether indicate that CB waste indirectly affect key adult life traits of Aedes aegypti and could conceivably be developed as a novel dengue vector control strategy, referring to previously documented direct toxicity on the larval stage. But this will require further research on CB waste effects on non-target organisms including humans.

Are exotic invaders less susceptible to native predators? A test using native and exotic mosquito species in New Zealand

A major hypothesis in invasion ecology is that native predators have less impact on exotic relative to native prey species. Here, we tested this hypothesis by examining the New Zealand native predator Anisops wakefieldi consuming native (Culex pervigilans) and exotic (Aedes notoscriptus) mosquito larvae. Anisops wakefieldi exhibited a decelerating type II functional response for both prey species, but at high prey densities consumed more of the exotic mosquito Ae. notoscriptus. A significantly higher attack rate was observed for the native predator feeding on exotic species. In the presence of both prey species, the predator showed preferences towards Ae. notoscriptus and demonstrated switching behavior towards this exotic species. The preference of the native predator towards the exotic mosquito appeared related to behavioral differences between the two prey species. We tested the behavioral response of both mosquito species in four conditions; (1) control (without predators), (2) free-roaming predators, (3) caged predators, and (4) kairomones only. Resting activities at the water surface and wall positions were the most frequently behaviors exhibited by Cx. pervigilans in the presence of predator. In contrast, the exotic species Ae. notoscriptus demonstrated significantly higher levels of “thrashing” behaviors, apparently making itself more obvious to the predators. The behaviors showed by Cx. pervigilans fulfilled the “threat sensitivity hypothesis”. No evidence here supported the idea that exotic mosquito species are less prone to the influence of native predators and the “escape from natural enemies” hypothesis seems not apply in our study. In fact, we observed that this invader was more susceptible to the predator.

DISCARDED CIGARETTE BUTTS ATTRACT FEMALES AND KILL THE PROGENY OF AEDES ALBOPICTUS

Abstract Discarded cigarette butts (DCB) waste occurs worldwide, pollutes landscapes, is unsightly, and results in added debris removal costs. There is, therefore, a great deal of current interest in making use of DCBs in beneficial ways. Despite evidence that DCBs are harmful to water fleas (Daphnia magna), which breed in aquatic environments as do mosquito larvae, their impact on dengue vectors is unknown. We examined whether Aedes albopictus alters its ovipositional responses, larval eclosion, and development in response to presence of DCBs in its habitats. We found oviposition activity in DCB-treated water similar to that of control water and that ovipositional activity in DCB solutions steadily increased over time as those solutions aged to 10 days. Larval eclosion was initially suppressed on day 1 in DCB solution, but increased thereafter to levels similar to control larval eclosion rates. The DCB–water solutions produced significantly higher mortality in both 1st and 2nd instars over control larvae for several days after initial exposure. Mortality rates decreased sharply 3 to 5 days postexposure as DCBs continued to decompose. We found increased survival rates during late development, but daily input of fresh DCBs prevented most young larvae from completing development. Taken together, these observations suggest that decomposing did not deter gravid Ae. albopictus females from ovipositing in treated containers and that DCB solutions had larvicidal effects on early instars. Our results are discussed in the context of DCB use to control container-breeding Ae. albopictus, a competent dengue vector in Asia and other parts of the world.

Larvicidal Efficacy of Different Plant Parts of Railway Creeper, Ipomoea cairica Extract Against Dengue Vector Mosquitoes, Aedes albopictus (Diptera: Culicidae) and Aedes aegypti (Diptera: Culicidae)

ABSTRACT. Natural insecticides from plant origin against mosquito vectors have been the main concern for research due to their high level of eco-safety. Control of mosquitoes in their larval stages are an ideal method since Aedes larvae are aquatic, thus it is easier to deal with them in this habitat. The present study was specifically conducted to explore the larvicidal efficacy of different plant parts of Ipomoea cairica (L.) or railway creeper crude extract obtained using two different solvents; methanol and acetone against late third-stage larvae of Aedes albopictus (Skuse) and Aedes aegypti (L.) (Diptera: Culicidae). Plant materials of I. cairica leaf, flower, and stem were segregated, airdried, powdered, and extracted using Soxhlet apparatus. Larvicidal bioassays were performed by using World Health Organization standard larval susceptibility test method for each species which were conducted separately for different concentration ranging from 10 to 450 ppm. Both acetone and methanol extracts showed 100% mortality at highest concentration tested (450 ppm) after 24 h of exposure. Results from factorial ANOVA indicated that there were significant differences in larvicidal effects between mosquito species, solvent used and plant parts (F = 5.71, df = 2, P < 0.05). The acetone extract of I. cairica leaf showed the most effective larvicidal action in Ae. aegypti with LC50 of 101.94 ppm followed by Ae. albopictus with LC50 of 105.59 ppm compared with other fractions of I. cairica extract obtained from flower, stem, and when methanol are used as solvent. The larvae of Ae. aegypti appeared to be more susceptible to I. cairica extract with lower LC50 value compared with Ae. albopictus (F = 8.83, df= l, P <0.05). Therefore, this study suggests that the acetone extract of I. cairica leaf can be considered as plant-derived insecticide for the control of Aedes mosquitoes. This study quantified the larvicidal property of I. cairica extract, providing information on lethal concentration that may have potential for a more eco-friendly Aedes mosquito control program.

Turning cigarette butt waste into an alternative control tool against an insecticide-resistant mosquito vector

Annually, 4.5 trillion cigarette butts (CBs) are flicked into our environment. Evidence exists that CB waste is deadly to aquatic life, but their lethality to the aquatic life of the main dengue vector is unknown. CBs are full of toxicants that occur naturally, during planting and manufacturing, which may act as larvicidal agents. We assessed Aedes aegypti vulnerability to Marlboro butts during its development. Overall, CBs showed insecticidal activities against larvae. At early phases of development, mortality rates were much higher in two CBs solution (2CBSol) and 3CBSol microcosms (MICRs). Larval survival gradually decreased with development in 1CBSol-MICRs. However, in great presence of CBs, mortality was high even for the late developmental stages. These results suggest that A. aegypti larvae are vulnerable to CB presence in their habitats, but this effect was seen most during the early developmental phases and in the presence of increased amounts of cigarette remnants. CB filters are being used as raw material in many sectors, i.e., brick, art, fashion, plastic industries, as a practical solution to the pollution problem, the observed butt waste toxicity to mosquito larvae open new avenues for the identification of novel insecticide products.

Occurrence of a mosquito vector in bird houses: Developmental consequences and potential epidemiological implications

Even with continuous vector control, dengue is still a growing threat to public health in Southeast Asia. Main causes comprise difficulties in identifying productive breeding sites and inappropriate targeted chemical interventions. In this region, rural families keep live birds in backyards and dengue mosquitoes have been reported in containers in the cages. To focus on this particular breeding site, we examined the capacity of bird fecal matter (BFM) from the spotted dove, to support Aedes albopictus larval growth. The impact of BFM larval uptake on some adult fitness traits influencing vectorial capacity was also investigated. In serial bioassays involving a high and low larval density (HD and LD), BFM and larval standard food (LSF) affected differently larval development. At HD, development was longer in the BFM environment. There were no appreciable mortality differences between the two treatments, which resulted in similar pupation and adult emergence successes. BFM treatment produced a better gender balance. There were comparable levels of blood uptake and egg production in BFM and LSF females at LD; that was not the case for the HD one, which resulted in bigger adults. BFM and LSF females displayed equivalent lifespans; in males, this parameter was shorter in those derived from the BFM/LD treatment. Taken together these results suggest that bird defecations successfully support the development of Ae. albopictus. Due to their cryptic aspects, containers used to supply water to encaged birds may not have been targeted by chemical interventions.

Risky behaviors: effects of Toxorhynchites splendens (Diptera: Culicidae) predator on the behavior of three mosquito species.

Viable biocontrol agents for mosquito control are quite rare, therefore improving the efficacy of existing biological agents is an important study. We need to have a better understanding of the predation-risk behavioral responses toward prey. This research examined prey choices by Toxorhynchites splendens by monitoring the behavioral responses of Aedes aegypti, Aedes albopictus, and Anopheles sinensis larvae when exposed to the predator. The results show that Tx. splendens prefers to consume Ae. aegypti larvae. The larvae exhibited different behavioral responses when Tx. splendens was present which suggest vulnerability in the presence of predators. “Thrashing” and “browsing” activities were greater in Ae. aegypti larvae. Such active and risky movements could cause vulnerability for the Ae. aegypti larvae due to increasing of water disturbance. In contrast, Ae. albopictus and An. sinensis larvae exhibited passive, low-risk behaviors, spending most of the time on the “wall” position near the edges of the container. We postulated that Ae. aegypti has less ability to perceive cues from predation and could not successfully alter its behavior to reduce risk of predation risk compared with Ae. albopictus and An. sinensis. Our results suggest that Tx. splendens is a suitable biocontrol agent in controlling dengue hemorrhagic vector, Ae. aegypti.
