The Use of Lewis Acid in the Reaction of Zinc Salts of Indoles and Acyl Chloride

Chengxi Yang; Hemantkumar H. Patel; Yi-Yin Ku; Rajendra Shah; David P. Sawick

Abstract 3-Acylindoles have been prepared in 62 – 92% isolated yield from the corresponding indoles and acyl chlorides in the presence of AlCl3.

Tetrahedron Letters | 1993

Cleavage of geminal diacetates using various phenoxides

Yi-Yin Ku; Ramesh R. Patel; David P. Sawick

Abstract Potassium phenoxides (phenoxide, 4-fluorophenoxide, 3-dimethylamino phenoxide) were found to cleave various geminal diacetates and regenerate aldehydes in excellent yields under extremely mild conditions.

Tetrahedron Letters | 1995

A practical synthesis of chiral propargylic alcohols

Yi-Yin Ku; Ramesh R. Patel; E.Michael Elisseou; David P. Sawick

Abstract A practical and efficient synthesis of chiral propargylic alcohols has been developed starting from inexpensive and readily available chiral methyl lactate. In addition, a regioselective desilylation method was discovered for oxygen deprotection of bis-silylated alkynols having a terminal trimethylsilyl group.

Tetrahedron Letters | 1994

Synthesis of substituted heterocycles. Simple method for the introduction of the N-hydroxyurea functionality

Yi-Yin Ku; Ramesh R. Patel; Brian A. Roden; David P. Sawick

Abstract A simple method for the introduction of the N-hydroxyurea functionality has been developed. This involves conjugate addition of hydroxylamine to a vinylsulfone substrate. This method was successfully used in a facile synthesis of the 5-lipoxygenase inhibitors, zileuton and A-69412.

SYNTHESIS OF A-79175 : A SECOND GENERATION 5-LIPOXYGENASE INHIBITOR

Daniel A. Dickman; Yi-Yin Ku; Howard E. Morton; Sanjay R. Chemburkar; Hemantkumar H. Patel; Albert V. Thomas; Daniel J. Plata; David P. Sawick

Abstract A convergent, high yielding, and scalable synthesis of A-79175, with a key step involving a mild and efficient CuPd catalyzed coupling reaction of a terminal acetylene with a substituted 2-iodofuran is discussed.

Synthesis of the macrolide antibiotic, oxolide

Brian A. Roden; Sanjay Chemburkar; Leslie A. Freiberg; Yi-Yin Ku; Richard J. Pariza

Abstract Oxolide was synthesized from Erythromycin A. Key features include two stereoselective reductions with DIBALH and NaBH3CN and a novel pH dependent purification of the final product. An acid-stable macrolide, oxolide, was synthesized from Erythromycin A. Key features include two stereoselective reductions with DIBALH and NaBH3CN and a simple pH dependent separation of the final product from its C-11 epimer.
