Biliary and urinary metabolites of estrone in the guinea pig.

Abstract The biliary and urinary metabolites of [6,7-3H]-estrone in guinea pigs with biliary fistulas were analyzed. In the bile, estrone-3-glucosiduronate was a major metabolite; and estrone-3-sulfate and polar conjugates (probably, diglucosiduronates and disulfates) were minor metabolites. In the urine, estrone-3-glucosiduronate and 17β-estradiol-3-glucosiduronate were detected. Moreover, polar conjugates (probably diglucosiduronates) were also found, but estrone-3-sulfate could not be detected. The possibility exists that estrone-3-glucosiduronate is derived from the liver and is partly converted to 17β-estradiol-3-glucosiduronate extrahepatically.

The metabolism of estrone and estrone-3-glucosiduronate in the guinea-pig

Abstract Analysis of urinary metabolites of [6,7- 3 H]-estrone and of [6,7- 3 H]-estrone-3-glucosiduronate was performed in intact guinea-pigs. The preliminary separation of urinary metabolites was carried out by submitting these metabolites to countercurrent distribution. The further separation of each fraction thus obtained was achieved by means of DEAE-Sephadex A-25 column chromatography. The separated peak was then hydrolyzed with the enzyme and the free steroid released was identified by reverse isotope dilution. Thus, both estrone-3-glucosiduronate and 17β-estradiol-3-glucosiduronate were identified as major urinary metabolites of estrone and of estrone-3-glucosiduronate. Concomitant studies in vitro with homogenates of the kidney, small intestine and liver showed that the small intestine and liver readily conjugated estrone as glucosiduronates and the kidney was a rather poor conjugating organ. The kidney and liver were capable of forming estrone-3-glucosiduronate as well as 17β-estradiol-3-glucosiduronate, whereas estrone-3-glucosiduronate was the only glucosiduronate formed by the small intestine. From these results, it was concluded that in this species not only the liver but the small intestine and kidney participated in the metabolic conjugation of estrone and that a part of estrone-3-glucosiduronate was converted to 17β-estradiol-3-glucosiduronate extrahepatically at least in the kidney.

Metabolic clearance rate of aldosterone and the electrolyte and water metabolism in hepatic disorders

The peculiar i t ies of re ta ining BSP were invest igated in hepatic diseases, especially for the purpose to s tudy the pathogenesis of const i tu t ional hyperbi l i rubinemia. Methods and Subjects: Total (T), free (F) and conjugated (C) BSP levels were examined by chromatography before and 15 minutes a f te r in t ravenous inject ion of dehydrocholic acid (DHC) of l g , which were performed immediately a f te r collecting a blood sample at 45 minutes in routine BSP test . The subjects included 16 l iver cirrhosis, 18 hepat i t is , 22 obstruct ive jaundice and 4 const i tut ional hyperbi l i rubinemia, diagnosed by biopsy, operat ion or autopsy. Results: a) In l iver cirrhosis, F-BSP decreased, but C-BSP showed li t t le changes af ter DHC injection. b) In hepat i t is , T-BSP and F-BSP showed no uni form changes. Elevat ion of C-BSP was observed in th ree cases with pers is tent hepat i t i s . c) In ex t rahepat ic obstruct ive jaundice, depression of F-BSP and elevat ion of C-BSP were observed af te r the injection. In in t rahepat ic obstruct ive jaundice, the changes in T-BSP, F-BSP and C-BSP were not uniform, but C-BSP/F-BSP rat io was h igher t h a n tha t in extrahepat ic . d) In const i tu t ional hyperbi l i rubinemia, Dubin-Johnsons syndrome showed s t r ik ing elevat ion of T-BSP, F-BSP and C-BSP af ter the inject ion. The elevation was more marked than spontaneous elevation observed character is t ical ly in this disease af ter rout ine BSP test . On the contrary in Rotor s hyperbi l i rubinemia, T-BSP and F-BSP showed dis t inct depression af ter the injection.

異なった核種の放射性ヨウ素で標識した蛋白を同時に投与することによって, 同一患者の同一時点における蛋白代謝を比較しようとした。すなわち, 125I-標識アルブミン, 131I-標識γ-グロブリンを正常, 肝硬変症, ネフローゼ症候群, malabsorption syndrome患者に同時に静注し, また, 2種の131I, 125I-標識パラプロテインを形質細胞腫患者に投与して, 125I, 131Iを分離測定することにより, その血漿放射能消失関係をもとめ, その数学的解析をおこなった。125Iおよび131Iは, 化学的分離操作をおこなうことなく, liquid scintillation spectrometerの波高分析回路とwell typeシンチレーション検出器とを組合わせた装置を用いて測定操作によって分離した。
