Differentiation of Herpes Simplex Virus Type 1 and Type 2 by Intranuclear Tubular Structures in Clinical Infection

Tubular structures (TS) or filamentous structures found in herpes simplex virus (HSV)-infected cells were first reported by Murphy et al (7) in mouse brains infected with HSV, and since then many authors have reported that these structures are found only in HSV type 2 (HSV-2)-infected cells, not in HSV type 1 (HSV-I)-infected cells (1,2,8, 12, 14). However, to our knowledge, there have been no reports dealing with many fresh isolates except for those of Nii and coworkers (9, 10); all other reports dealt with only one to three strains of HSV-2. The study reported here was undertaken to determine the role of TS as a morphological marker in HSV-2 infection by examining 17 newly isolated strains of HSV (9 strains of HSV-I and 8 strains of HSV-2). Viruses were isolated from 15 patients at the Department of Dermatology of Kyushu University Hospital. The patients, diagnoses, and sites oflesions are listed in Table 1. Strains Nos. 4 and 5, and Nos. 9 and 10 were obtained from the same patients, respectively. All the specimens were collected with sterile swabs during the vesicular stage of infection and the swabs were dipped in 1 ml of Eagles minimum essential medium supplemented with 2% calf serum (CS), 40 units of penicillin per ml and 100 ftg of streptomycin per ml. Vero cell monolayers in culture bottles were infected with the specimens. When a maximum cytopathic effect (CPE) was observed, the culture fluid containing infected cells was frozen, thawed, and then sonically treated. Cell debris was removed by centrifugation at 3,000 rpm for 15 min. The virus-containing supernatant fluid was stored at -70 C. Two laboratory strains, KOS (HSV-I) and YS-4 (HSV-2) (6), were used as controls and maintained in the same manner as described above. Type-specific antiserum was prepared by the method of Glorioso et al (3). White rabbits were injected intraperitoneally with the KOS or YS-4 strain of HSV six times at weekly intervals and hyperimmune serum was collected 7 days after the last injection. The serum was absorbed with lyophilized rabbit kidney cells (RK-13 line) infected with heterotypic HSV. Neutralizing antibody titers of absorbed sera were 16 for homotypic viruses and less than 2 for heterotypic viruses. Biological Properties of Herpes Simplex Virus Type 2 Isolated from the Recurrent Facial Lesions of an Infant

生後4～5日目に初発し, 3年間に計8回顔面皮疹の再発をくりかえした患者の顔面病巣から5株の2型単純ヘルペスウイルスが分離された。この患者の再発性の皮疹が毎回同じ性状のウイルスによつて起こつているか否かを知る目的で, 分離された5株のヘルペスウイルスについて生物学的性状, 血清学的反応, および電子顕微鏡観察による比較を行ないつぎの結果を得た。 1) 分離された5株はいずれも単純ヘルペスウイルス2型であつた。 2) ニワトリ胎児初代培養細胞におけるプラックサイズ, 発育鶏卵漿尿膜上でのポックサイズをマーカーとした生物学的性状については5株の間に有意の差はみられなかつた。また成熟マウスにたいする神経毒性も5株の間に大きなちがいはなかつた。 3) 抗1型および抗2型単純ヘルペスウイルス血清による中和反応でも5株の間に大きなちがいはなかつた。
