Intraperitoneal Alpha-Lipoic Acid to prevent neural damage after crush injury to the rat sciatic nerve

Objective Crush injury to the sciatic nerve causes oxidative stress. Alfa Lipoic acid (a-LA) is a neuroprotective metabolic antioxidant. This study was designed to investigate the antioxidant effects of pretreatment with a-LA on the crush injury of rat sciatic nerve. Methods Forty rats were randomized into four groups. Group I and Group II received saline (2 ml, intraperitoneally) and a-LA (100 mg/kg, 2 ml, intraperitoneally) in the groups III and IV at the 24 and 1 hour prior to the crush injury. In groups II, III and IV, the left sciatic nerve was exposed and compressed for 60 seconds with a jewelers forceps. In Group I (n = 10), the sciatic nerve was explored but not crushed. In all groups of rats, superoxide dismutase (SOD) and catalase (CAT) activities, as well as malondialdehyde (MDA) levels were measured in samples of sciatic nerve tissue. Results Compared to Group I, Group II had significantly decreased tissue SOD and CAT activities and elevated MDA levels indicating crush injury (p < 0.05). In the a-LA treatment groups (groups III and IV), tissue CAT and SOD activities were significantly increased and MDA levels significantly decreased at the first hour (p < 0.05) and on the 3rd day (p < 0.05). There was no significant difference between a-LA treatment groups (p > 0.05). Conclusion A-LA administered before crush injury of the sciatic nerve showed significant protective effects against crush injury by decreasing the oxidative stress. A-LA should be considered in the treatment of peripheral nerve injuries, but further studies are needed to explain the mechanism of its neuroprotective effects.

Effects of early administration of dexamethasone, N-acetylcysteine and aprotinin on inflammatory and oxidant–antioxidant status after lung contusion in rats

INTRODUCTION This experimental setting was undertaken to elucidate and confirm the role of inflammatory and oxidant-antioxidant mechanisms on blunt injury induced moderate pulmonary contusion (PC). We intended to determine the effects of dexamethasone (DXM), N-acetylcysteine (NAC) and aprotinin (APR) in terms of their ability to diminish the consequences of acute lung injury due to PC. METHODS Rats were allocated to five subgroups. Except for the control, all subgroups had a moderate pulmonary contusion. Following 45 min of observation, animals in groups I and II received intraperitoneal saline, group III 10 mg/kg DXM, group IV 500 mg/kg NAC and group V 30,000 kIU/ml APR. After the procedure, 6 h after contusion, blood gas analysis, lung tissue nitric oxide (NO) and malondialdehyde (MDA) levels, superoxide dismutase (SOD) and catalase (CAT) activity, bronchoalveolar lavage (BAL) fluid and histopathological examination were performed. RESULTS All PaO(2) values decreased significantly in contused rats as compared with the control group (p<0.05). DXM, NAC and APR resulted in a slight increase in PaO(2) values compared with group II (p<0.05). Lung tissue levels of MDA and NO were higher in the contusion group than in the control (p<0.05). DXM, NAC and APR all decreased the levels of MDA and NO (p<0.05), however the decrease in NO was not found to be significant with APR (p>0.05). SOD and CAT activities increased significantly after contusion compared to control group (p<0.05). There was no significant difference even though SOD levels were elevated in groups III, IV and V compared with contused animals (p>0.05). Neutrophils in BAL fluid significantly increased in contused animals (p<0.05). Only DXM significantly decreased neutrophil population in BAL fluid (p<0.05). Scores for alveolar haemorrhage/oedema were higher in all contusion-performed rats than those in the control (p<0.05). Compared with the other drugs, only APR significantly improved the haemorrhage/oedema scores compared to sham animals (p=0.024). CONCLUSIONS Our findings demonstrate that moderate bilateral PC induced by blunt chest trauma leads to an early inflammatory process which is clearly associated with activation of the oxidant-antioxidant cascade. On this basis, early supportive treatment with DXM, NAC and APR may yield favourable results on pulmonary pathophysiological parameters which are adversely affected due to PC.

Morphometrical features of the human malleolar groove.

Background: Our aim was to investigate the morphometric features of malleolar groove of the lateral malleolus in humans. Methods: This study was performed on the malleolar grooves of 93 (80 dry and 13 cadaveric) fibulae. We measured the length of the fibula; length, width, and depth of the malleolar groove; the angle between malleolar groove and the fibular axis. Furthermore, we measured the distances between the distal tip of the fibula and the lateral and medial tubercles of lateral malleolus, and the width of posterior surface of the lateral malleolus. Results: We found the shape of malleolar groove regularly concave in 63 out of 93 (68%) fibulae. The features of the triangular area at the posterior aspect of the lateral malleolus, where the peroneal tendons pass over, may serve as a bed for the peroneal tendons. There was a significant correlation between the depth of the malleolar groove and the distances between the distal tip of the fibula and the medial and lateral tubercles, as well (p < 0.05). Conclusion: A strong correlation existed between either the shape of the groove or the values of the reference points. Clinical Relevance: The shape of the groove may be important in reinforcing the superior peroneal retinaculum (SPR) and other structures restraining the tendons.

The comparative investigation of morphology of papillary muscles of left ventricle in different species

The papillary muscles of the heart are conical projections into the respective chamber of myocardium covered by endocardium. Functionally, the papillary muscles are important part of the respective valve complex. They prevent the cusps of a valve from being everted when the ventricle contracts. Our study was conducted on papillary muscles in left ventricle of 135 normal adult hearts (20 humans, 25 dogs, 60 sheep and 30 goats). The length (L), width (W), head number (HN), angle with ventricular wall (A°), distance with annulus (AnD) and apex (ApD), and the shapes of the anterior and posterior papillary muscles were observed. The tendinous chords (CTN) attached to each papillary muscle were counted at their origin. The values of both anterior and posterior papillary muscles were not statistically significant (p > 0.05) in measurements of L, W, HN, A° and AnD of all species, but those of CTN and ApD were significant (p < 0.05) between human and animal hearts, while the difference was not significant (p > 0.05) between sheep and goat hearts. Knowledge of morphology of papillary muscles in different species will be of much use to clinical investigators in their studies.

Effect of Atorvastatin and Ezetimibe Treatment on Serum Lipid Profile and Oxidative State in Rats Fed With a High-Cholesterol Diet

Introduction:The effects of ezetimibe and atorvastatin on serum lipid profile and oxidant-antioxidant system were investigated in rats. Methods:Seventy-two Sprague-Dawley rats were assigned to 6 groups. Group 1 was fed with standard rat chow. Group 2 and the other 4 groups were fed with a high-cholesterol diet: 10 mg/kg/d atorvastatin to group 3; 1 mg/kg/d atorvastatin to group 4; 10 mg/kg/d atorvastatin and 1 mg/kg/d ezetimibe to group 5; and 1 mg/kg/d ezetimibe to group 6. After 3 months, serum total, low-density lipoprotein, high-density lipoprotein cholesterol, and triglyceride levels, and the activities of malondialdehyde, glutathione peroxidase, and superoxide dismutase were measured in the plasma. In addition, the left anterior descending and femoral arteries were examined histopathologically. Results:Serum total, low-density lipoprotein cholesterol, and triglyceride levels decreased slightly in group 3. However, administration of 1 mg/kg/d atorvastatin or 1 mg/kg/d ezetimibe did not significantly change lipid parameters. Plasma malondialdehyde levels slightly increased in group 2 compared with controls and decreased compared with both the atorvastatin regimens. However, malondialdehyde levels increased with the addition of ezetimibe to atorvastatin. Only the administration of ezetimibe significantly elevated the levels of malondialdehyde. Glutathione peroxidase and superoxide dismutase levels were also found to be significantly reduced in the groups receiving ezetimibe when compared with atorvastatin groups. Conclusions:Atorvastatin has a beneficial effect on oxidative stress in rats fed with high-cholesterol diet. A combination of ezetimibe with atorvastatin diminishes the beneficial effects of atorvastatin. Conversely, the sole administration of ezetimibe increases oxidative stress.

The histopathological investigation of the effect on regional and systemic tissues of the application of Medicinal Plant Extract Ankaferd Blood Stopper in deep tissue injuries in Rats

BACKGROUND This study was planned to evaluate both the histopathological changes under light microscope as well as the systemic organ effects following application of Ankaferd Blood Stopper® (ABS) (a mixture of five plant extracts) in an animal model of deep tissue hemorrhage. METHODS A total of 50 Wistar Albino rats were divided into five groups of 10 rats each. The rats underwent femoral vein puncture and were treated with ABS tampon, ABS spray, or Surgicel, and one group was left untreated. After two weeks, each group underwent partial tissue excision from the same femoral region as well as from the brain, heart, kidney, and liver. RESULTS The specimens from all groups were obtained from the femoral region after two weeks and evaluated under light microscope. The light microscope revealed no histopathological changes in neurovascular structures or in deep connective tissues in any of the groups. CONCLUSION ABS provided hemostasis and was observed to stop bleeding. There were no histopathological changes at the tissue level and no pathological effects in other organs tissues under light microscope, and the remote organ tissue remained clear.

Comparison of transforming growth factor β1 concentrations in the ovaries of rats stimulated by human menopausal gonadotropin or recombinant follicle-stimulating hormone

OBJECTIVE To compare the effects of human menopausal gonadotropins (hMG) and recombinant follicle stimulating hormone (rFSH) on transforming growth factor (TGF) β1 concentration in the rat ovary. STUDY DESIGN Twenty-one fertile Wistar-Albino rats were divided into 3 groups of 7. Groups 1, 2 and 3 were injected with saline, hMG or rFSH, respectively, over 5 days, after which they underwent ovariectomy. Hematoxylin and eosin (H&E) staining was used for histological examination. TGF β1 staining levels in ovarian stroma, vessel walls, granulosa cells of Graafian follicles and corpus luteum cells were investigated immunohistochemically. RESULTS On histological examination, the number of smaller antral follicles was higher in the control group, while there were more and larger antral follicles in the hyperstimulated groups. There were statistically significant differences in staining in vessel walls and granulosa cells between the control and stimulated groups. Both stimulation protocols caused an increased TGF β1 concentration in vessel walls, while there was weak staining in granulosa cells in the treatment groups compared to the control group (p<0.05). There were no significant differences in staining scores between the two treatment groups (p>0.05). CONCLUSIONS The effects of two different gonadotropin preparations on TGF β1 concentrations in different localizations in the rat ovaries are comparable. It may be postulated that the luteinizing hormone (LH) content of hMG contributes little or nothing to the TGF β1 mediated angiogenesis.

Analysis in terms of gender of morphometric characteristics of typical cervical vertebrae: A radiological study -

The objective of this study is to examine the morphometric characteristics of C3-C6 vertebrae in the cervical region through radiological images and to find out how they differ in terms of gender. CT images of 27 men and 27 women between the ages of 18 and 40 who did not have any symptoms were included in the study. In the study, area of vertebra (AV), vertebral foramen (VF), right and left transvers foramen (RTF-LTF) areas, lateral diameter of vertebral foramen (VF-L) and anterior-posterior diameter of vertebral foramen (VF-AP) of 216 cervical typical vertebrae (C3-C6) were measured through radiological images. No statistically significant results were found between AV, VF, VF-L, VF-AP, RTF and LTF radiological measurements taken from the C3, C4, C5 and C6 vertebrae of men and women for the same vertebral levels and between the rates of VF, RTF and LTF areas covered in AV (p>0.05). Paired comparisons of vertebrae were conducted with independent samples t test. An anatomic overview to manspreading campaign

There is a campaign called “manspreading”. The campaign which started to draw attention to men’s making passengers uncomfortable by sitting with open legs in public transportation, attracted too much attention. When we glance through communal living spaces, we see men mostly sitting with their legs open. Can there be a morphological background of this sitting position? Basically, the pelvis of men and women are different from each other. Angulus subpubicus is bigger in women. In women, the femur head is located to lateral due to excess angulus subpubicus. However, the lower end of femur has to direct to medial to adjust with tibial condyle, that is, it gets closer to midline. Anteversion angle of acetabulum is higher in women. Anteversion with a higher angle means more anteriorly located acetabulum. That is, both femurs have to be more located to the midline. In women, the collodiaphyseal angle and femur neck length is smaller than men. Both of these situations cause femurs to be located close to the midline. Q angle which is used in the assessment of the mechanic and situation of knee joint is higher in women. High Q angle means that femur lower ends are close to the midline. Due to anatomical differences listed above, women’s femurs are located closer to the midline, while the femurs of men are located more laterally. This difference is also reflected in the way they sit.

The role of chrysin against harmful effects of formaldehyde exposure on the morphology of rat fetus liver and kidney development

This study was aimed to investigate possible harmful effects of formaldehyde (FA) exposure on the morphology of fetus liver and kidney development during pregnancy and also to determinate possible protective role of chrysin (CH) against these harmful effects. For this aim, after pregnancy was induced, 58 female rats were divided into 6 groups. Serum physiologic (SF) was injected to the Group I rats intraperitoneally (i.p.). 20 mg/kg CH was given to the Group II via gavage. 0.1 mg/kg FA was applied to the Group III (i.p.), 1 mg/kg FA was injected to Group IV (i.p.) 0.1 mg/kg FA was given to Group V i.p., and 20 mg/kg CH was given to the same group via gavage. 1 mg/kg FA was applied to Group VI i.p., and 20 mg/kg CH was given to the same group via gavage. Fetuses were taken from each pregnant rat with cesarean section on the 20th day of the pregnancy. The morphological analyses of the fetuses, liver and kidney; biochemical and histological analyses of the liver and kidney were performed. The fetal body, liver and kidney weight of the FA groups demonstrated a statistically significant decrease the compared to control group. Also the FA-1 group were observed histopathological changes on the fetus liver and kidneys. FA exposure causes harmful effects on fetus the liver and kidneys. CH reduces the negative effect on morphological variables statistically. Although CH is insufficient to fix the histopathological changes that occur in the liver, damaging effects that occur in the kidney decreased statistically.
