Determination of dehydroepiandrosterone, androsterone, etiocholanolone, and “IIβ-hydroxyetiocholanolone” in plasma

Abstract A method for the estimation of dehydroepiandrosterone, androsterone, etiocholanolone, and “IIβ-hydroxyetiocholanolone” in 10–20 ml of plasma is described. For cleavage of conjugates in plasma extracts, solvo lysis with subsequent hot acid hydrolysis of non-solvolysable conjugates is employed. Separation of 17-ketosteroids is achieved by paper chromatography in propylene glycol-methylcyclohexane, the single fractions being determined by a micro zimmermann reaction. Plasma levels of the four 17-ketosteroids in a few normal adults indicate a rather wide range of the individual concentrations.

Percutane Katheterisierung der rechten und linken Nebennierenvene beim Menschen

Zusammenfassung1. Es wird eine Methode der percutanen Katheterisierung beider Nebennierenvenen mitgeteilt. Die Katheterisierung der rechten und der linken Nebennierenvene erfolgt nacheinander in einem Untersuchungsgang von einer Punktionsstelle aus am nüchternen, nicht sedierten Patienten.2. Das hierzu erforderliche Instrumentarium, insbesondere die benötigten Katheter werden beschrieben.3. Bei 40 Patienten wurde die Methode im Verlauf einer endokrinologischen Diagnostik und bei 10 Patienten mit nicht dekompensierten angeborenen Herzfehlern während einer Herzkatheterisierung angewandt. Es gelang bei den insgesamt 50 Patienten in 3 Fällen nicht, die rechte und in 6 Fällen nicht, die linke Nebennierenvene zu katheterisieren. Zwischenfälle und Komplikationen traten nicht auf.4. Bei den 10 endokrinologische unauffälligen Patienten mit angeborenen Herzfehlern lagen die Cortisolkonzentrationen des rechten Nebennierenvenenplasmas im Mittel 10mal und die des linken 14mal höher als im peripheren Plasma, wo die Konzentrationen zwischen 4,6 und 18,6 µg Cortisol/100 ml variierten.5. Untersuchungen der 17-OH-CS-Ausscheidung im Harn und der 17-OH-CS-Konzentrationen im Plasma vor, während und nach der Katheterisierung ergaben keinen Hinweis auf eine wesentliche Veränderung der Nebennierenrindeninkretion als Folge des Untersuchungsganges.6. Die Methode erscheint für eine spezielle endokrinologische Diagnostik geeignet. Die Ergebnisse der Cortisolbestimmungen im Nebennierenvenen- und peripheren Plasma werden im Hinblick auf die Cortisol-Sekretionsrate diskutiert.Summary1. A method is described for the successive percutaneous catheterization of both adrenal veins via the right femoral vein under local anesthesia.2. The catheterization instruments consist of radiopaque teflon catheters, usual stainless steel spring guides and specially curved stiff guides.3. Catheterization of the adrenal veins was performed in 40 patients under study for endocrine disorders and in 10 patients with congenital heart disease without failure in the course of a routine cardiac catheterization. Catheterization of adrenal veins was unsuccessful 3 times on the right and 6 times on the left side. All catheterization were uneventful.4. The adrenal and peripheral venous concentrations of cortisol were determined in the 10 cardiac patients without endocrine disorders. Cortisol concentrations of the right and left adrenal venous plasma averaged 10 and 14 times the peripheral venous plasma concentrations of 4.6–18.6 µg/100 ml.5. The catheterization procedure did not cause a consistent change of adrenal cortical function as judged by determination of the urinary 17-OH-CS excretions before, during and after catheterization. In two patients studied the peripheral venous plasma 17-OH-CS concentrations were not increased in the course of catheterization. A normal decrease of plasma 17-OH-CS concentration during daytime which had occurred previously under normal conditions was not observed on the day of catheterization.6. The method is considered to be suitable for special endocrinological studies. The adrenal venous cortisol concentrations are discussed.

Bestimmung von Oestrogenen durch Densitometrie ihrer ABS- oder DANSYL-Derivate

Oestrogens have been reacted with azobenzene-4-sulphonic acid chloride (ABS-chloride) or 1-1-dimethylaminonaphthyl-5-sulphonyl chloride (DANSYL-chloride) in alkaline aqueous acetonic solution and the derivatives obtained (yield about 95%) have been separated by thin-layer chromatography on silica gel. Quantitative measurement has been performed by densitometry or fluorimetry.ZusammenfassungDurch Reaktion von Oestrogenen mit Azobenzol-4-sulfonsäurechlorid (ABS-Chlorid) oder 1-Dimethylaminonaphthyl-5-sulfonylchlorid (DANSYL-Chlorid) in alkalischer, wäßrig-acetonischer Lösung wurden die entsprechenden Derivate hergestellt (Ausbeute etwa 95%), die anschließend durch Dünnschicht-Chromatographie auf Kieselgel getrennt wurden. Die quantitative Messung erfolgte densitometrisch oder fluorimetrisch.

Chromatography of uronic acids, sugar acid lactones, and glucuronosides on anion-exchange paper or DEAE-cellulose.

Abstract For the separation of uronic acids, sugar acid lactones, and glucuronosides paper chromatography on anion-exchange paper, such as Whatman ET-20, Whatman DE-20, and Schleicher & Schull Anionenaustauscher Papier with Dowex 2 X8 may be employed. Mixtures of alcohols, water, and organic acids or buffers were as mobile phases. In a similar manner, glucuronosides and glucuronic acid or glucuronic acid γ-lactone can be successfully chromatographed on columns of anion-exchange cellulose (Whatman DE-50 floc).

Estimation of glucuronosides by paper chromatography

Abstract A method for the quantitative estimation of glucuronosides in urine is described. Glucuronosides are separated by paper chromatography of purified urinary extract on Ecteola anion exchange paper (Whatman ET-20) or by column chromatography on DEAE cellulose (Whatman DE-50 floc). Glucuronosides present in paper or column eluates are estimated by the naphthoresorcinol reaction. Concentrations of glucurono- sides in urine as determined by this method are considerably lower than those reported for other procedures not involving purification or separation of the material to be estimated.

Estimation of pregnanediol and pregnanetriol in urine

Abstract A method for the simultaneous determination of pregnanediol and pregnanetriol in urine has been developed. It consists of the enzymatic hydrolysis of conjugated steroids by β-glucuronidase, extraction, purification of extracts, spread-layer chromatography, and quantitation by well known color reactions. The accuracy of the method and its precision can be considered adequate, as judged by recovery experiments and multiple analyses. The sensitivity of the method lies at 2.5 μg pregnanediol and 0.8 μg pregnanetriol per sample. The specificity of the procedure depends on spread-layer chromatography. Due to the rapidity and efficiency of the chromatographic step the applicability of the method may be regarded satisfactory. Levels of pregnanediol and pregnanetriol in urine of normal adult subjects lie well in the range observed by estimation with other methods.

Über steroid-konjugate in plasma III. Quantitative bestimmung unpolarer 17-ketosteroid-konjugate in menschlichem plasma

Zusammenfassung Bei Chromatographie eines Plasmaextraktes an Aluminiumoxyd fanden sich 80% der gesamten 17-KS in lipoid-haltigen Fraktionen, wahrend nur 20% in ihrem chromatographischen Verhalten 17-KS-sulfaten entsprachen. Nachfolgende Chromatographie des unpolaren Materials an Kieselsaure fuhrte zu einer betrachtlichen Verringerung der unpolaren 17-KS-konjugate bei Zunahme der sich wie 17-KS-sulfate verhaltenden Fraktion. Das Gemisch unpolarer 17-KS-konjugate enthielt Dehydroepiandrosteron, Androsteron, Aetiocholanolonund “IIβ-Hydroxyaetiocholanolon”.

Direct metabolism of 7α-3H-DHEA-3-14C-glucuronoside

Es wird gezeigt, dass beim Menschen Steroide in Form ihrer 3-Glucuronoside metabolisiert werden können, und zwar in ähnlicher Weise wie dies schon für entsprechende 17-Glycoside nachgewiesen worden ist.

Bei der in-vivo Perfusion menschlichen Lungengewebes mit 7α-3H-DHEA-35S-sulfat zeigte es sich, dass im Lungengewebe sowohl Sulfatase- wie auch Sulfokinaseaktivität enthalten ist. Als Metaboliten konnten neben dem vorherrschenden 5-Androsten-3Β, 17Β-diol auch Testosteron, Androstendion, Androsteron, ätiocholanolon sowie Oestrogene nachgewiesen werden.

Enterohepatischer Kreislauf von 7α-3H-DHEA-14C-Glukuronosid und 7α-3H-DHEA-35S-Sulfat beim Meerschweinchen

By infusing doubly labelled conjugates into the duodenum or into a mesenteric vein it could be demonstrated that DHEA-sulfate passes the intestinal wall as well as the liver nearly without hydrolysis. However, most of the glucuronoside is hydrolysed in the intestine and reconjugated as sulfate.
