An experiment (complete randomised design) was conducted to investigate the effects of Laminaria hyperborea and Laminaria digitata seaweed extract inclusion on gut morphology, selected intestinal microbiota populations, volatile fatty acid concentrations and the immune status of the weaned pig. Twenty-eight piglets (24 days of age, 6.5 ± 1.4 kg live weight) were assigned to one of four dietary treatments for 7 days and then sacrificed: (T1) basal diet (control); (T2) basal diet and 1.5 g/kg L. hyperborea seaweed extract; (T3) basal diet and 1.5 g/kg L. digitata seaweed extract; and (T4) basal diet and 1.5 g/kg of a combination of L. hyperborea and L. digitata seaweed extract. The seaweed extract contained both laminarin and fucoidan. Digesta samples were taken from the caecum and colon to measure the enterobacteria, bifidobacteria and lactobacilli populations and for volatile fatty acid analysis. Tissue samples were taken from the duodenum, jejunum and ileum for morphological examination. Blood samples were taken to determine the cytokine gene expression profile and to measure the phagocytotic capacity of the blood. Pigs offered diets containing L. hyperborea seaweed extract had less bifidobacteria in the colon (P < 0.05) and lactobacilli in the caecum (P < 0.05) and colon (P < 0.001). The inclusion of L. digitata seaweed extract resulted in lower populations of enterobacteria in the caecum and colon (P < 0.01), bifidobacteria in the caecum (P < 0.05), and lactobacilli in the caecum (P < 0.05) and colon (P < 0.001). Pigs offered the combination of L. hyperborea and L. digitata seaweed extracts had less enterobacteria (P < 0.05) and lactobacilli (P < 0.01) in the caecum and colon. Pigs offered the L. digitata-supplemented diet had a reduced villous height in the duodenum and jejunum (P < 0.05). The inclusion of the L. digitata seaweed extract increased the molar proportion of butyric acid in the colon (P < 0.05). There was a significant reduction in the ammonia concentration in the colon with the inclusion of L. hyperborea (P < 0.01) and L. digitata (P < 0.05) seaweed extracts. An increase in the expression of the Interleukin-8 mRNA was observed on day 6 with the supplementation of the combination of L. hyperborea and L. digitata seaweed extract (P < 0.05). The inclusion of L. hyperborea seaweed extract resulted in an increase in total monocyte number (P < 0.05). In conclusion, the supplementation of L. hyperborea and L. digitata seaweed extract alone and in combination reduced the enterobacteria, bifidobacteria and lactobacilli populations in the caecum and colon, while only marginal effects on the immune response was observed.

A 2 × 2 factorial experiment was conducted to investigate the interaction between cereal type (wheat v . barley) and an exogenous enzyme supplement (with or without) on nutrient digestibility, large intestinal microflora, volatile fatty acid profile and in vitro manure ammonia emissions from finisher pigs. The enzyme supplement used contained endo-1, 3-β-glucanase (EC 3·2·1·6) and endo-1, 4-β-xylanase (EC 3·2·1·8). The diets were formulated to contain similar concentrations of net energy (9·8 MJ/kg) and lysine (10·0 g/kg). Urine and faeces were collected over seven consecutive days from 16 boars (four boars per treatment, 80·0 kg live weight) that were housed in metabolism crates. After collections, the pigs were slaughtered and the contents of the intestinal tracts were removed for analysis. There was a significant interaction between cereal type and enzyme inclusion in the apparent total tract digestibility of dry matter (DMD), organic matter (OMD) and nitrogen. The inclusion of an enzyme supplement in barley-based diets increased ( P P in vitro ammonia emissions. Twenty piglets (21 days, 7·8 kg live weight (LW)) were used in a 2×2 factorial to investigate interactions between lactose and inulin on intestinal morphology, microbiology and volatile fatty acid (VFA) production of the weanling pig. The piglets were offered the following diets for 6 days and then sacrificed: (T1) 150 g/kg lactose, (T2) 150 g/kg lactose +15 g/kg inulin, ( T3) 330 g/kg lactose, and ( T4) 330 g/kg lactose +15 g/kg inulin. Tissue samples were taken from the duodenum, jejunum and ileum for morphological measurements. Digesta samples were taken from the ileum, caecum and colon. BACKGROUND In experiment 1, 30 boars were assigned to one of five treatments (n = 6): T1, 0 g kg(-1) seaweed extract (SWE); T2, 0.7 g kg(-1) SWE; T3, 1.4 g kg(-1) SWE; T4, 2.8 g kg(-1) SWE and T5, 5.6 g kg(-1) SWE. The extract contained laminarin and fucoidan only and was extracted from Laminaria spp. In experiment 2, 28 boars were assigned, in a 2 x 2 factorial to one of four treatments (n = 7): T1, control; T2, control plus 300 mg laminarin; T3, control plus 240 mg fucoidan; T4, control plus 300 mg laminarin and 240 mg fucoidan kg(-1) diet. RESULTS In experiment 1 there was a response to SWE on colonic Bifidobacterium spp. (P < 0.01 quadratic), Enterobacterium spp. (quadratic P < 0.05) and on caecal Enterobacterium spp. (quadratic P < 0.05). In experiment 2 there was an interaction (P < 0.05) between laminarin and fucoidan supplementation on Enterobacterium spp. in the proximal and distal colon. Pigs offered laminarin had reduced Enterobacterium spp. compared with pigs offered the control diet. However, the combination of laminarin and fucoidan had increased Enterobacterium spp. compared with alone. Pigs offered diets containing fucoidan had increased Lactobacilli spp. in the proximal colon (P < 0.05) and distal colon (P < 0.001) compared with non-fucoidan diets. CONCLUSION Overall, the reductions in intestinal Enterobacterium spp. and increases in Lactobacilli spp. obtained suggest that laminarin and fucoidan may provide a dietary means to improve gut health in pigs.

An experiment (complete randomised design) was conducted to investigate the linear and quadratic effects of barley β-glucan inclusion level on total tract nutrient apparent digestibility, nitrogen excretion, intestinal microflora, volatile fatty acid (VFA) profile and manure ammonia emissions in pigs. Twenty-four boars (66 kg) were assigned to one of four treatments: (T1) 0 g/kg barley (control diet) (5.6 g/kg β-glucan), (T2) 222 g/kg barley (12.1 g/kg β-glucan), (T3) 444 g/kg barley (18.9 g/kg β-glucan) and (T4) 666 g/kg barley (25 g/kg β-glucan). Barley was substituted for wheat in the diet. The diets were formulated to contain similar concentrations of digestible energy and digestible lysine. There was a linear decrease (P < 0.001) in the total tract apparent digestibility of dry matter, organic matter, gross energy and neutral-detergent fibre with increasing β-glucan concentration. Faecal nitrogen excretion was affected by dietary β-glucan concentration (quadratic P < 0.05). There was a linear decrease in Enterobacteria concentrations (P < 0.05) with increasing dietary β-glucan concentration. Increasing dietary barley levels caused a linear decrease in colonic (P < 0.01) and caecal pH (P < 0.001). Total caecal VFA and propionic acid were affected by dietary β-glucan concentration (quadratic, P < 0.05). There was a linear decrease in the proportion of acetic acid (P < 0.001), isobutyric acid (P < 0.01) and isovaleric acid (P < 0.05) with increasing levels of dietary barley in both the caecum and colon. There was a linear increase in the proportion of propionic acid (P < 0.001) and butyric acid (P < 0.05) with increasing barley in the colon. In conclusion, high level of dietary β-glucan (25 g/kg) is required to reduce offensive odour forming branched-chain VFAs; however, diet digestibility is compromised at such levels.

A 2 × 2 factorial experiment was conducted to investigate the interaction between high and low dietary crude protein (CP) (200 v. 150 g/kg) and sugar-beet pulp (SBP) (200 v. 0 g/kg) on nutrient digestibility, nitrogen (N) excretion, intestinal fermentation and manure ammonia and odour emissions from 24 boars (n = 6, 74.0 kg live weight). The diets were formulated to contain similar concentrations of digestible energy (13.6 MJ/kg) and lysine (10.0 g/kg). Pigs offered SBP-containing diets had a reduced (P < 0.05) digestibility of dry matter, ash, N, gross energy and an increased (P < 0.001) digestibility of neutral-detergent fibre compared with pigs offered diets containing no SBP. There was an interaction between CP and SBP on urinary N excretion and the urine : faeces N ratio. Pigs offered the 200 g/kg CP SBP-based diet had reduced urine : faeces N ratio (P < 0.05) and urinary N excretion (P < 0.05) compared with those offered the 200 g/kg CP diet without SBP. However, there was no effect of SBP in pigs offered 150 g/kg CP diets. Manure ammonia emissions were reduced by 33% from 0 to 240 h (P < 0.01); however, odour emissions were increased by 41% (P < 0.05) when pigs were offered SBP diets. Decreasing dietary CP to 150 g/kg reduced total N excretion (P < 0.001) and ammonia emissions from 0 to 240 h (P < 0.05). There was an interaction between dietary CP and SBP on branched-chain fatty acids (P < 0.001) in caecal digesta. Pigs offered the 200 g/kg CP SBP-containing diet reduced branched-chain fatty acids in the caecum compared with pigs offered the 200 g/kg CP diet containing no SBP. However, there was no effect of SBP in the 150 g/kg CP diet. In conclusion, pigs offered SBP-containing diets had a reduced manure ammonia emissions and increased odour emissions compared with diets containing no SBP. Pigs offered the 200 g/kg CP SBP-containing diet had a reduced urine : faeces N ratio and urinary N excretion compared with those offered the 200 g/kg CP diet containing no SBP.

This experiment sought to establish the response to increasing levels of coconut oil (CO) supplementation with a fixed 0·50:0·50 forage:concentrate diet on intake, digestibility and methane (CH 4 ) emissions. Sixteen continental cross beef heifers (mean starting weight 481±36 kg) were assigned randomly to one of four levels of CO; 0 g/day, 125 g/day, 250 g/day or 375 g/day in an incomplete (three periods) multiple (no. =4) Latin-square design experiment (no. =12 per treatment). A linear reduction in CH 4 output occurred as the level of CO in the diet increased ( P P 4 output was greater than the proportional reduction in DMI and hence CH 4 l/kg DMI decreased from 39·8 l/kg when no CO was given to 29·7 l/kg when 375 g/day CO was given. The addition of CO to the diet resulted in a significant decline in dry-matter digestibility (DMD) at the 375 g/day level ( P 4 production with no adverse effect on DMI or DMD up to the 250 g/day level (0·027 of dietary DM).

One hundred and eighty piglets (24 days old, 6·0 kg live weight) were used to investigate interactions between lactose, avilamycin and inulin on growth performance and nutrient digestibility in weaned piglets. The piglets were blocked on the basis of live weight and within each block assigned to one of six dietary treatments (six replicates (pens) per treatment). The piglets were offered diets containing either a low (175 g/kg) or high (295 g/kg) lactose levels with one of the following food additives (1) 0 supplementation (2) avilamycin (60 mg/kg) or (3) inulin (15 g/kg) in a 2 × 3 factorial arrangement. The objective of the current experiment was to evaluate the influence of dietary cereal sources of beta(1,3)(1,4)-d-glucan (beta-glucan) and enzyme supplementation on indices of environmental pollution from finisher pigs. An experiment with a 2 x 2 factorial arrangement of treatments was initiated to investigate the effect of dietary source of beta-glucan (barley vs. oats) and enzyme supplementation (no vs. yes) on nutrient digestibility, N utilization, intestinal fermentation, and manure odor and ammonia emissions from finisher boars (n = 4; BW = 73.9 kg; SD = 4.7). Sixteen boars were assigned to 1 of 4 dietary treatments (n = 4/treatment): 1) barley-based diet, 2) barley-based diet + exogenous enzyme, 3) oat-based diet, and 4) oat-based diet + enzyme. The enzyme supplement used contained endo-1,3(4)-beta-glucanase and endo-1,4-beta-xylanase. Experimental diets were formulated to contain similar concentrations of DE (13.5 MJ/kg) and digestible lysine (8.8 g/kg). Pigs offered oat-based diets had reduced digestibility of DM (0.795 vs. 0.849; SEM 0.007; P = 0.001), OM (0.808 vs. 0.865; SEM 0.007; P = 0.001), GE (0.806 vs. 0.845; SEM 0.006; P = 0.002), and NDF (0.233 vs. 0.423; SEM 0.033; P < 0.003) compared with those offered barley-based diets. Oat-based diets increased populations of Bifidobacterium spp. (7.26 vs. 6.38 log cfu/g of digesta; SEM 0.201; P = 0.005) and Lactobacillus spp. (6.99 vs. 6.18 log cfu/g of digesta; SEM 0.234; P = 0.022) in the proximal colon and decreased manure odor emissions [2,179.6 vs. 4,984.6 Ou(E)/m(3) (where Ou(E) refers to European odor units); SEM 653.7; P < 0.011] compared with barley-based diets. There was an interaction between cereal type and enzyme inclusion on manure ammonia emissions from 0 to 96 h (P = 0.050). Pigs offered barley-based diets containing an enzyme supplement had increased manure ammonia emissions compared with those offered unsupplemented barley-based diets. However, there was no effect of enzyme inclusion on oat-based diets. In conclusion, pigs offered oat-based diets harbored increased Bifidobacterium spp. and Lactobacillus spp. populations in the proximal colon and had decreased manure odor emissions compared with those offered barley-based diets. Enzyme inclusion had no effect on manure ammonia emissions from pigs offered oat-based diets.

Two experiments were conducted to investigate the use of dietary manipulation as a means of improving piglet post-weaning performance and gastro-intestinal health. In experiment 1, 144 piglets (24 days old) in a 3 × 2 factorial arrangement were offered diets containing 65, 170 and 280 g lactose per kg with or without lactic acid (16 g/kg) for 28 days. In experiment 2, 20 piglets (24 days old) (in a 2 × 2 factorial arrangement were offered the following diets for 7 days and then sacrificed: T1) basal diet; T2) basal diet + 15 g inulin per kg; T3) basal diet + 16 g lactic acid per kg and T4) basal diet + 15 g inulin per kg + 16 g lactic acid per kg. After slaughtering, tissue samples were taken from the duodenum, jejunum and ileum for morphological measurements. Digesta samples were taken from the ileum, caecum and colon for microbiology and volatile fatty acid analysis. In experiment 1, pigs offered diets containing lactic acid had improved daily gain ( P P P P P P P E. coli ( P P E. coli concentrations compared with the inulin only diets ( P E. coli numbers in isolation of the other. In conclusion, in experiment 1, lactic acid improved performance in the 1st week post weaning. There was a linear increase in ADG with increasing lactose levels. In experiment 2, the combination of lactic acid and inulin increased villous height in the jejunum and concentrations of lactobacilli and E. coli in the colon.
