Evaluation of sugar content and composition in commonly consumed Korean vegetables, fruits, cereals, seed plants, and leaves by HPLC-ELSD

In the present investigation, evaluation of sugars viz. fructose, galactose, glucose, sucrose, lactose, maltose, and raffinose in commonly consumed raw Korean vegetables, fruits, cereals, seed plants, and leaves has been analyzed using high-performance liquid chromatography with evaporative light scattering detection (HPLC-ELSD). Of the 58 samples analyzed, Onion showed the highest content of fructose (27.74g/100g) and glucose (31.80g/100g) and Chestnuts showed the highest content of sucrose (21.82g/100g). On the other hand, Glutinous sorghum (polished grain), Green tea leaves, and Paddy rice (well-polished rice) showed the lowest content of fructose (0.20g/100g), glucose (0.68g/100g) and sucrose (0.23g/100g), respectively. Glutinous barley (Hopimbori-whole grain) and Green tea leaves showed 0.17g/100g and 0.57g/100g of galactose and lactose respectively. Glutinous barley (Seodunchalbori-polished grain) and Black soybeans (Cheongjaho, dried) showed the highest content of maltose (0.51g/100g) and raffinose (1.82g/100g), respectively. In few samples, galactose, maltose, lactose, and raffinose were detected in trace quantities. A partial least squares discriminant analysis (PLS-DA) was also performed to discriminate the analyzed samples.

Effects of in ovo injection with selenium on immune and antioxidant responses during experimental necrotic enteritis in broiler chickens

This study was conducted to investigate the effects of in ovo injection of Se on modulating the immune system and antioxidant responses in broiler chickens with experimental necrotic enteritis. Broiler eggs were injected at 18 d of embryo age with either 100 μL of PBS alone or sodium selenite (Na2SeO3) in PBS, providing 0 (SS0), 10 (SS10), or 20 (SS20) μg of Se/egg. At 14 d posthatch, PBS-treated and uninfected chickens were kept as the control group, whereas the remaining chickens were orally infected with 1.0 × 10(4) sporulated oocysts of Eimeria maxima (SS0, SS10, SS20). At 18 d posthatch, E. maxima-infected chickens were orally infected with 1.0 × 10(9) cfu of Clostridium perfringens. Infected control SS0 group showed significantly decreased BW compared with the uninfected control. However, SS20 group showed significantly increased BW compared with the infected control SS0 group, whereas the BW were similar among uninfected control and infected SS10 and SS20 groups. The SS10 group showed significantly lower intestinal lesions compared with the SS0 group, and oocyst production was decreased in both SS10 and SS20 groups. Serum malondialdehyde level and catalase activity were also decreased in both SS10 and SS20 groups, whereas the superoxide dismutase level was significantly lower in the SS10 group compared with the SS0 group. The SS20 group showed significantly higher levels of transcripts for IL-1β and IL-6 in intestine, and SS10 and SS20 groups had higher levels of transcripts for IL-8 and inducible nitric oxide synthase expression and decreased glutathione peroxidase 7 mRNA levels compared with the SS0 group. The SS10 and SS20 groups also showed increased serum antibody levels to C. perfringens α-toxin and NetB toxin compared with the SS0 group. These collective results suggest that the injection of Se into the amniotic cavity of developing eggs may be beneficial for enhancing immune and antioxidant responses in the hatched chickens exposed to the necrotic enteritis-causing pathogens.

Estrogen-like activity of aqueous extract from Agrimonia pilosa Ledeb. in MCF-7 cells

BackgroundPostmenopausal women experience estrogen deficiency-related menopausal symptoms (e.g., hot flashes and mood swings) and a dramatic increase in the incidence of chronic diseases. Although estrogen-replacement therapy (ERT) can reduce mortality from cardiovascular disease and improve osteoporosis and menopausal symptoms, its side effects have limited recent use. This study investigated the estrogen-like activity of aqueous extract from Agrimonia pilosa Ledeb.MethodsThe estrogenic activity of A. pilosa was investigated by using several in vitro assays. The binding activity of A. pilosa on estrogen receptors was examined using a fluorescence polarization-based competitive binding assay. The proliferative activity of A. pilosa was also examined using MCF-7 cells. Furthermore, the effect of A. pilosa on the expression of 3 estrogen-dependent genes was assessed.ResultsUsing liquid chromatography-mass spectrometry, the 3 major peaks of A. pilosa aqueous extract were identified as apigenin-hexose, luteolin-glucuronide, and apigenin-glucuronide. The aqueous extract induced the proliferation of estrogen receptor-positive MCF-7 cells (p < 0.05). A. pilosa-stimulated proliferation was blocked on adding the estrogen antagonist ICI 182,780. Moreover, A. pilosa treatment increased the mRNA expression of the estrogen-responsive genes pS2 and PR (p < 0.05).ConclusionsThese results suggest A. pilosa can be used to improve estrogen deficiency-related menopausal symptoms or to treat diseases in postmenopausal women.

Hog millet (Panicum miliaceum L.)-supplemented diet ameliorates hyperlipidemia and hepatic lipid accumulation in C57BL/6J-ob/ob mice.

Dietary intake of whole grains reduces the incidence of chronic diseases such as obesity, diabetes, cardiovascular disease, and cancer. In an earlier study, we showed that Panicum miliaceum L. extract (PME) exhibited the highest anti-lipogenic activity in 3T3-L1 cells among extracts of nine different cereal grains tested. In this study, we hypothesized that PME in the diet would lead to weight loss and augmentation of hyperlipidemia by regulating fatty acid metabolism. PME was fed to ob/ob mice at 0%, 0.5%, or 1% (w/w) for 4 weeks. After the experimental period, body weight changes, blood serum and lipid profiles, hepatic fatty acid metabolism-related gene expression, and white adipose tissue (WAT) fatty acid composition were determined. We found that the 1% PME diet, but not the 0.5%, effectively decreased body weight, liver weight, and blood triglyceride and total cholesterol levels (P < 0.05) compared to obese ob/ob mice on a normal diet. Hepatic lipogenic-related gene (PPARα, L-FABP, FAS, and SCD1) expression decreased, whereas lipolysis-related gene (CPT1) expression increased in animals fed the 1% PME diet (P < 0.05). Long chain fatty acid content and the ratio of C18:1/C18:0 fatty acids decreased significantly in adipose tissue of animals fed the 1% PME diet (P < 0.05). Serum inflammatory mediators also decreased significantly in animals fed the 1% PME diet compared to those of the ob/ob control group (P < 0.05). These results suggest that PME is useful in the chemoprevention or treatment of obesity and obesity-related disorders.

Evaluation of γ-oryzanol content and composition from the grains of pigmented rice-germplasms by LC-DAD-ESI/MS

BackgroundRice is the staple food and one of the world’s three major grain crops. Rice contains more than 100 bioactive substances including phytic acid, isovitexin, γ-oryzanol, phytosterols, octacosanol, squalene, γ-aminobutyric acid (GABA), tocopherol, tocotrienol derivatives, etc. Out of them, γ-oryzanol is known to have important biological profile such as anti-oxidants, inhibitor of cholesterol oxidation, reduce serum cholesterol levels in animals, effective in the treatment of inflammatory diseases, inhibit tumor growth, reduce blood pressure and promotes food storage stability when used as a food additive, etc. Hence in the present investigation, we aimed to evaluate the content and composition of γ-oryzanol from pigmented rice germplasms using a liquid chromatography with diode array detection and electrospray ionization-mass spectrometry (LC-DAD-ESI/MS).FindingsIn the present study, 33 exotic pigmented rice accessions (red, white and purple) have been evaluated. Among them, the contents of γ-oryzanol varied from 3.5 to 21.0Âmg/100Âg with a mean of 11.2Âmg/100Âg. A total of ten components of γ-oryzanol including ∆7-stigmastenyl ferulate were identified of which, cycloartenyl ferulate, 24-methylenecycloartanyl ferulate, campesteryl ferulate and sitosteryl ferulate were identified as the major components. The mean proportions of steryl ferulates were in the descending order of 24-methylenecycloartanyl ferulate > cycloartenyl ferulate > campesteryl ferulate > sitosteryl ferulate > ∆7-campestenyl ferulate > campestanyl ferulate > sitostanyl ferulate > ∆7-stigmastenyl ferulate > stigamsteryl ferulate > ∆7-sitostenyl ferulate. Almost 11 accessions (33%) showed higher content than the control rice Chucheongbyeo and higher proportions ranged from 10 to 15Âmg/100Âg. Interestingly, the red rice accession Liberian Coll. B11/B-11 (21.0Âmg/100Âg) showed higher content γ-oryzanol than control rice Jeokjinjubyeo (19.1Âmg/100Âg) and the purple rice accession Padi Adong Dumarat, Mardi No.4376 (20.3Âmg/100Âg) showed a similar content with control rice Heugjinjubyeo (21.4Âmg/100Âg).ConclusionsMost of analyzed rice accessions were found to possess higher contents of γ-oryzanol than the control rice, Chucheongbyeo. In particular, the red accessions showed highest content than the white and purple accessions. The content and composition of γ-oryzanol in 33 exotic pigmented rice accessions have been evaluated and compared significantly by the present investigation.

Antioxidant activities and determination of phenolic compounds isolated from oriental plums (Soldam, Oishiwase and Formosa).

The purposes of this study were to determine phenolic compounds and to evaluate antioxidant activities of plums (Soldam, Oishiwase and Formosa). Soldam contains the highest amount of total phenolics among cultivars (Formosa: 4.0%, Oishiwase: 3.3%, Soldam: 6.4% for total phenolic) as well as the total flavonoids of which constituents were mainly myricetin and anthocyanidin. The antioxidant activities were measured by DPPH, ABTS radical scavenging, and SOD-like activities. The DPPH radical scavenging activity of Korean plum extracts (200 µg/mL) showed more than 43%, and the Soldam turned out to be the highest : ID50 value: 160-177 µg/mL for Formosa and Oishiwase; 58-64 µg/mL for Soldam. The ABTS radical scavenging activity of Korean plum extracts (200 µg/mL) was found to be more than 50%. The SOD-like activity of Korean plum extracts (200 µg/mL) showed more than 70%. Among three kinds of cultivars, Soldam had the highest antioxidant activity. The nitrite scavenging activity of Soldam was 61.5%, which is the highest, compared with that of the other cultivars, about 50%. From these results, Korean plums turned out to be phytochemical rich fruit as well as to show high antioxidant activities.

Characterization and quantification of γ-oryzanol in grains of 16 Korean rice varieties

Abstract γ-Oryzanol, a mixture of ferulic acid esters of triterpene alcohols and sterols, is a nutritionally important group of rice secondary metabolites. A library of 27 γ-oryzanol was assembled from existing data and used to assist identification and quantification of γ-oryzanol isolated from 16 Korean rice varieties (11 white and 5 pigmented). γ-Oryzanol was analyzed with liquid chromatography with diode array detection and electrospray ionization mass spectrometry. Nineteen different γ-oryzanol were observed and identified as stigmasterol, campesterol and sitosterol or common and hydroxylated triterpene alcohols. In the 16 varieties, the total γ-oryzanol content averaged 43.8 mg/100 g (range, 26.7–61.6 mg/100 g), which Josaengheugchal exhibited the highest level (61.6 mg/100 g). The Korean rice varieties were classified based on qualitative and quantitative γ-oryzanol data by multivariate statistical analysis. Clusters of specialty rice varieties exhibited higher γ-oryzanol levels than those of common rice varieties.

In Vitro Analysis of the Immunomodulating Effects of Allium Hookeri on Lymphocytes, Macrophages, and Tumour Cells

The present study investigated the effects of ethanol extracts of Allium hookeri (leaf, root, and fermented root) on parameters of innate immunity, tumour cell viability and antioxidant effect in vitro. Innate immunity was measured by spleen lymphocyte proliferation, nitric oxide production by chicken macrophage HD11 cells and suppressive effect on tumour cell viability was assessed using chicken RP9 cells. Free radical scavenging capacity as a measure of antioxidant capacity was determined by 0.15 mM of DPPH solution. In vitro culture of chicken spleen lymphocytes with ethanol extract of Allium hookeri (62.5–500 µg/mL) significantly induced higher proliferation compared with media control. Stimulation of macrophages with ethanol extract of Allium hookeri (62.5–500 µg/mL) showed increased Nitric oxide production. Tumor cells growth was significantly inhibited by extracts of Allium hookeri at 15.6–125 µg/mL compared with medium control and all extracts exhibited greater than 80% scavenging activity at 1000 µg/mL compared with ethanol vehicle control. Above all, fermented root extracts showed strongest effects on antioxidant activity compared to leaf and root extracts.

Characterization and Quantification of Flavonoid Glycosides in the Prunus Genus by UPLC-DAD-QTOF/MS

Widely distributed in plants, flavonoids reduce the incidence of cancer and cardiovascular disease. In this study, flavonoid content and composition in members of the Prunus genus were evaluated using liquid chromatography with diode array and electrospray ionization mass spectrometric detection (UPLC-DAD-ESI/QTOF-MS). Flavonoids in plants of the Prunus genus include the basic structures of kaempferol, quercetin, and catechin, and exist as mono-, di-, or tri-glycoside compounds mono-acylated with acetic acid. A total of 23 individual flavonoids were isolated and confirmed, three of which appear to be newly identified compounds: quercetin 3-O-(2″-O-acetyl)neohesperidoside, quercetin 3-O-(4″-O-acetyl)rutinoside, and kaempferol 3-O-(4″-O-acetyl)rutinoside. Japanese apricot and Chinese plum contained the highest amounts of flavonoids in the Prunus genus. During the ripening stage of Japanese apricot, the total flavonol content was reduced, while the catechin content was increased.

Changes in Antioxidant and Antimicrobial Activities of Schizandra chinensis Baillon under Different Solvent Extraction

The present study was performed to investigate antioxidant and antimicrobial activities of Schizandra chinensis Baillon under different extraction conditions. Extraction yield of water extract, 70% ethanol extract, and methanol extract from S. chinensis was 17.7, 32.9, and 36.6% respectively. Total polyphenol content was 9.2-16.3 mg/g in S. chinensis extracts and was increased significantly by ethanol and methanol extraction (p < 0.05). Radical scavenging activity against DPPH and ABTS·+ was significantly higher in ethanol and methanol extracts than in water extract (p < 0.05). Ethanol and methanol extracts from S. chinensis also showed effective superoxide dismutase-like activity compared with water extract (p < 0.05). S. chinensis water extract exhibited strong antimicrobial activity against Escherichia coli, Staphylococcus aureus, and Salmonella typhimurium, which was increased significantly by ethanol and methanol extraction (p < 0.05). On the other hand, inhibitory effect of S. chinensis water extract on nitric oxide production was more effective than ethanol and methanol extract.
