Characterization of Fiber Types in Different Muscles of the Hindlimb in Female Weanling and Adult Wistar Rats

We analyzed lesser diameter and distribution of fiber types in different skeletal muscles from female Wistar rats using a histoenzymology Myofibrillar Adenosine Tri-phosphatase (mATPase) method. Fragments from muscles were frozen and processed by mATPase in different pH. Adult and weanling rat soleus muscles presented a predominance of type I fibers and larger fiber diameters. In the plantar muscle in adult rats, the type IIB fibers demonstrated greater lesser diameter while in the weanling animals, types I and IIB fibers were larger. The plantar muscle of animals of both ages was composed predominantly of the type IID fibers. The type IID fibers were observed in similar amounts in the lateral gastrocnemius and the medial gastrocnemius muscles. Type IIB fibers showed predominance and presented higher size in comparison with other types in the EDL muscle. The present study shows that data on fiber type distribution and fiber lesser diameter obtained in adult animals cannot always be applied to weanling animals of the same species. Using the mATPase, despite the difficult handling, is an important tool to determine the different characteristics of the specific fibers in the skeletal muscle tissue.

Propriedades mecânicas do gastrocnêmio eletroestimulado pós-imobilização

Introduction: Mechanical properties (MP) are clinically applicable tools for healthcare professionals working on the musculoskeletal system. Objectives: The aim of this study was to evaluate two protocols of neuromuscular electric stimulation (NMES) to improve MP regeneration of the myotendinous complex after segment immobilization in female rats. Materials and Methods: Fifty animals were equally distributed into five groups: Control (CG, n=10); Immobilized (IG, n=10); Immobilized and freely remobilized (IFG, n=10); Immobilized and NMES once /day (IEG1, n=10); Immobilized and MNES twice/day (IEG2, n=10). Immobilization was kept for 14 days, and remobilization was subsequently released for 10 days. NMES was applied for 10 days, post-immobilization, every morning for 10 minutes to IEG1 animals and every morning and afternoon (total 20 minutes) to the IEG2 group. After these procedures, the gastrocnemius muscle was submitted to the mechanical traction assay to evaluate stiffness, resilience, load and stretching at maximum limit MPs. Results: Immobilization reduced the MP values concerning load and stiffness (p 0.05). Conclusion: It is concluded that the gastrocnemius muscle became structurally better organized through a single NMES application and by remobilization.

Estimulação elétrica neuromuscular e o alongamento passivo manual na recuperação das propriedades mecânicas do músculo gastrocnêmio imobilizado

We evaluated the influence of immobilization, free remobilization, remobilization with manual passive stretching, remobilization with neuromuscular electric stimulation (NMES) and remobilization with electric stimulation and associated passive stretching on some mechanical properties of the gastrocnemius muscle of female rats. Sixty female rats were assessed, being distributed into 6 experimental groups. One of these groups served as control. The animals of the five remaining groups had their right posterior limb immobilized for 14 consecutive days. From the five groups, one was sacrificed right after the immobilization period, a second group was released from immobilization, a third was submitted to the manual passive stretching technique for 10 consecutive days, a fourth was submitted to NMES for 10 consecutive days and the last one was submitted to NMES and manual passive stretching for 10 consecutive days. We found that the immobilization caused a significant reduction of the mechanical properties values evaluated on the muscle. The free remobilization could not reestablish any of the properties. The remobilization by manual passive stretching restored the mechanical properties of stretching at the proportionality limit, stiffness and resilience. The remobilization stimulated by NMES reestablished all of studied properties. The remobilization by electric stimulation and passive stretching reestablished the mechanical properties of stretching at the maximum limit, proportionality limit, and stiffness.

Propriedades mecânicas do músculo gastrocnêmio de ratas, imobilizado e posteriormente submetido a diferentes protocolos de alongamento

Stretching is widely employed in physiotherapeutic clinical practice and in sportive activities; however, the mechanical alterations of the skeletal muscle generated by this technique are poorly scientifically investigated. This study evaluated the mechanical alterations suffered by the gastrocnemius muscle of young adult female Wistar rats, submitted to14 days of immobilization followed by passive manual stretching during 10 consecutive days once or twice a day. Fifty animals were equally distributed in five groups, Control (CG); Immobilized (IG); Immobilized and liberated (ILG); Immobilized and submitted to stretching once a day (IEG1); Immobilized and submitted to stretching twice a day (IEG2). The gastrocnemius muscle was analyzed by mechanical traction assay and the properties related to load and maximal and proportional stretching evaluated in addition to stiffness and resilience. Immobilization decreased load at maximal thresholds (MTL), load at proportional thresholds (LPT), stretch at maximal thresholds (SMT), stiffness and resilience were reduced in 44.4%, 34.4%, 27.6%, 64.4% and 54% respectively, compared to CG values. With subsequent free remobilization and stretching, all parameters were restored except for IEG2 in which SMT remained reduced in 31.3%, when compared to CG. It is concluded that after 14 days of segmental immobilization, individual stretching loads and free movements contribute to regain muscle mechanical properties.

Análise biomecânica e histológica de músculos gastrocnêmios de ratas submetidas à lesão muscular e tratados com laserterapia de baixa intensidade

OBJETIVE: To evaluate, by means of biomechanical and histological analysis, the effect of low-level laser therapy on the gastrocnemius of rats subjected to muscle lesion by impact. METHODS: 45 female Wistar rats, weighing 250g, were divided into 3 groups (n=15/group): C (control), ML (animals submitted to muscle lesion) and ML-L (animals submitted to muscle lesion and laser therapy). The animals from groups ML and ML-L were subjected to an experimental muscle lesion by means of an impact of a 250g load, released from a height of 30 cm, directly on the gastrocnemius. The animals from group ML-L were submitted to a treatment with a 960 Nm laser, 2 J/cm2, for 3 days, twice a day. RESULTS: The mechanical tests were performed on the Universal Testing Machine. The average values of maximum force were: 35.70 (± 2.69) N in group C, 31.77 (± 2.59) N in group ML and 34.36 (± 3.63) N in group ML-L, with a statistically significant difference between groups C and ML (p 0.05). Histological analysis showed a presence of blood vessels in group ML-L and a hematoma in the healing stage. CONCLUSION: Treatment with low-level laser had a positive effect in the healing process of muscle injury.

Efeitos morfológicos do retorno da sobrecarga após imobilização em alongamento de músculo esquelético de ratas

CONTEXTUALIZACAO: Na reabilitacao, a imobilizacao em alongamento do musculo esqueletico e realizada como contramedida para reverter efeitos de encurtamento muscular severo e em eventos pos-cirurgicos. Acredita-se que o retorno as atividades funcionais normais estimule mecanotransdutores capazes de reorganizar a citoarquitetura normal muscular, porem a descricao das alteracoes histopatologicas relacionadas a esses procedimentos sao escassas na literatura. OBJETIVOS: Avaliar e quantificar anomalias histologicas induzidas pela imobilizacao em alongamento do musculo EDL (Extensor Digitorum Longus) e confronta-las com a livre movimentacao do animal apos esse procedimento. METODOS: Foram utilizadas 18 ratas Wistar, distribuidas nos grupos: controle (GC); imobilizadas em flexao plantar (EDL em posicao alongada) por 14 dias (GI); imobilizadas por 14 dias e liberadas por dez dias (GIL). Fragmentos do EDL foram congelados, seccionados e processados com reacoes imuno-histoquimica para colagenos I e III e histoquimica para Adenosina Trifosfatase Miofibrilar e Hematoxilina-Eosina. RESULTADOS: Os animais do GI apresentaram discreto aumento da expressao de colageno I e de fibras em processo degenerativo/necrotico, reducao da proporcao de fibras tipo (FT) 2A e do diâmetro menor de todos os tipos de fibras, quando comparados com os animais do GC. Para o GIL, observou-se retorno da quantidade de colageno I as condicoes controle, alem de reducao na proporcao de FT2D, aumento do numero de nucleos centralizados e do diâmetro menor das fibras quando comparadas com o GI, porem a expressao de FT2B e FT2D nao atingiu os valores de referencia. CONCLUSOES: Os dados apresentados mostram que a retomada da funcao durante dez dias foi parcialmente eficiente na recuperacao das caracteristicas do musculo EDL apos o periodo de imobilizacao e que, se extrapolados os dados a clinica fisioterapeutica, a adocao de procedimentos orientados as disfuncoes primarias do musculo pode favorecer a resposta morfofuncional do segmento e o seu integro restabelecimento.

Role of Auriculotherapy in the Treatment of Temporomandibular Disorders with Anxiety in University Students

Introduction. The aim of this study was to evaluate the role of auriculotherapy with mustard seeds in the treatment of temporomandibular disorders (TMDs), anxiety, and electromyographic (EMG) activity in university students. Methodology. The State Trait Anxiety Inventory (STAI), Research Diagnostic Criteria (RDC) for TMDs (RDC/TMDs), and electromyography were used in this study of 44 college students with high levels of anxiety and TMDs. The subjects were divided into two groups: an auriculotherapy (AA) group (n = 31) and an AA sham group (n = 13). The mustard seeds were applied to the shenmen, rim, sympathetic, brain stem, and temporomandibular joint (TMJ) points in the AA group and to sham points in the external ear and wrist in the AA sham group. The treatment protocol was 10 sessions (two treatments per week). Results. Anxiety (p < 0.01) was significantly reduced in the AA group. This group also showed a decrease in tender points in the mandibular posterior region (p = 0.04) and in the right side of the submandibular region (p = 0.02). Complaints of bilateral pain were reduced in the temporal tendon (p ≤ 0.01) and in the left side of the ATM (p < 0.01). In addition, electromyographic (EMG) activity was reduced during temporal muscle contraction (p = 0.03).  Conclusion. Auriculotherapy was effective in the treatment of students with anxiety and TMDs.

Biomechanical and histological analysis of the gastrocnemius in rats subjected to muscle injury and treatment with low-level laser therapy

Objective: To mechanically and histologically evaluate the application of low-level laser therapy to the reparative process on lesions caused by impact on the gastrocnemius muscles of rats. Methods: 45 female Wistar rats were divided into three groups (n=15/ group): C (control, no lesion), ML (muscle lesion) and ML-L (muscle lesion and laser therapy). The experimental muscle lesion was produced by letting a 250 g load drop from a height of 30 cm, directly onto the muscle. The animals in the ML-L group were subjected to application of 960 nm laser, 2 J/cm2, on the lesion site, for three days, twice a day. Mechanical tests were performed on an Emic® universal testing machine. Results: The mean values for the maximum force were: 35.70 (± 2.69) N in group C, 31.77 (± 2.59) N in group ML and 34.36 (± 3.63) N in group ML-L, with a statistically significant difference between groups C and ML (p < 0.05). The mean values for relative stiffness were: 3.75 (± 0.98) N/mm in group C, 3.84 (± 0.32) N/mm in group ML and 4.43 (± 0.68) N/mm in group ML-L, with no statistically significant differences (p>0.05). Histological analysis showed the presence of blood vessels in group ML-L and hematomas during the repair process. Conclusion: Laser therapy had a positive effect on the regeneration process of the muscle injury.

Effect of Foot Reflexology on Capillary Blood Glucose, Tissue Temperature, and Plantar Pressure of Individuals With Diabetes Mellitus (Type 2): A Pilot Study

Objective The purpose of this study was to assess the effect of foot reflexology on capillary blood glucose, feet tissue temperature, and plantar pressure of the feet of individuals with diabetes mellitus (type 2). Methods Forty‐five individuals with type 2 diabetes mellitus were stratified into 2 groups: treated (n = 21), which received orientation about foot self‐care and received 12 foot reflexology sessions; and control (n = 24), which received only orientations about foot self‐care. A portable glucose meter, an infrared thermography camera, and a baropodometer evaluated the variables. Results The data indicate that, after 12 therapy sessions, there were no significant differences between the groups for producing effects on capillary blood glucose, feet tissue temperature, and plantar pressure. Conclusion No significant effect was observed after 12 foot reflexology sessions in the variables that were evaluated.

Effects of Exergaming in Cancer Related Fatigue in the Quality of Life and Electromyography of the Middle Deltoid of People with Cancer in Treatment: A Controlled Trial

Objective: In the present study, we aimed to evaluate the effects an exergaming protocol for cancer patients who undergo or have already undergone cancer treatment. We sought to evaluate changes in cancer-related fatigue, function, and ability to perform daily activities, in addition to changes in the electromyographic pattern of the middle deltoid muscle. Methods: We conducted a controlled trial. Nineteen volunteers in the cancer group (aged 61 ± 9 years; body mass index28 ± 5) and 19 in the control group (aged 58 ± 8 years); body mass index 28 ± 4) participated in the study. They were evaluated by means of a sociodemographic and clinical questionnaire, the Functional Assessment of Chronic Therapy-Fatigue (FACIT-F) questionnaire, and surface electromyography in the deltoid muscle at three moments: before the beginning of the exergaming protocol, after 10 training sessions, and after 20 sessions. The protocol consisted of practicing exergaming using Xbox 360® (Microsoft, Redmond, USA) with Kinect®. The game “Your Shape Fitness Evolved” (Ubisoft, Rennes, France) was used. Results: Total FACIT-F scores, fatigue subscale scores, and median frequency values observed in the cancer group were lower than those in the control group. These values improved in relation to the initial evaluation in the cancer group after the practice of the exergaming protocol. Conclusion: The exergaming protocol used in this study was effective for reducing reported symptoms of fatigue, increasing perceived quality of life, and improving the pattern of deltoid muscle contraction in cancer patients.
