Ultrasound-assisted extraction of D-pinitol from carob pods using Response Surface Methodology

This study reports on the optimization of the conditions of extraction of D-pinitol compound from carob pods by using ultrasonication. For this purpose, the Box-Behnken design, which is a widely used form of Response Surface Methodology, was used to investigate the effects of parameters on the ultrasound-assisted extraction. Four independent variables, temperature (°C), ultrasonic power (W), dilution rate (material:water ratio) and time (min), were studied. The results showed that the highest D-pinitol concentration of 11.98 g/L was obtained with an extraction temperature of 50 °C, ultrasonic power of 207 W, a dilution rate of 1:4 and an extraction time of 120 min. It was found that the D-pinitol concentration of carob extract increased with the effect of ultrasonication during the extraction process. Therefore, ultrasound-assisted extraction could be used as an alternative to conventional hot water extraction with respect to the recovery of D-pinitol from carob pods with the advantages of lower extraction temperatures, shorter extraction time and reduced energy consumption.

Effects of Baking and Boiling on the Nutritional and Antioxidant Properties of Sweet Potato [Ipomoea batatas (L.) Lam.] Cultivars

The effects of baking and boiling on the nutritional and antioxidant properties of three sweet potato cultivars (Beniazuma, Koganesengan, Kotobuki) cultivated in Turkey were investigated. The samples were analyzed for proximate composition, total phenolic content, ascorbic acid, β-carotene, antiradical activity, and free sugars. The dry matter, protein, and starch contents of the sweet potatoes were significantly changed by the treatments while the ash and crude fiber contents did not differ as significantly. The β-carotene contents of baked and boiled sweet potatoes were lower than those of fresh sweet potatoes; however, the total phenolic and ascorbic acid contents of the baked and boiled sweet potatoes were higher than those of the fresh samples. Generally, the antiradical activity of the sweet potatoes increased with the treatments. Sucrose, glucose, and fructose were quantified as free sugars in all fresh sweet potatoes; however, maltose was determined in the treated samples. In terms of the analyzed parameters, there were no explicit differences among the sweet potato cultivars.

Determination of D-pinitol in carob syrup.

Carob syrup is a traditional product native to the Mediterranean region, containing a high concentration of sugar, phenolic compounds and minerals. d-pinitol is a bioactive component extracted from legumes and has some beneficial effects on human metabolism. In this research, the d-pinitol content and sugar profile of 10 different carob syrup samples purchased from Turkish markets were determined. Mean d-pinitol, sucrose, glucose and fructose contents of samples were found to be 84.63 ± 10.73, 385.90 ± 45.07, 152.44 ± 21.72 and 162.03 ± 21.45 g/kg dry weight, respectively. Carob syrup has a considerable amount of d-pinitol compared with the other d-pinitol-including legumes. Consequently, this study showed that carob syrup may be a suitable source of d-pinitol for medical use and d-pinitol may be an indicator for the detection of any adulteration in carob syrup.

Ultrasound-assisted dilute acid hydrolysis of tea processing waste for production of fermentable sugar.

Lignocellulosic materials that are the most abundant plant biomass in the world have the potential to become sustainable sources of the produced value added products. Tea processing waste (TPW) is a good lignocellulosic source to produce the value added products from fermentable sugars (FSs). Therefore, the present study is undertaken to produce FSs by using ultrasound‐assisted dilute acid (UADA) and dilute acid (DA) hydrolysis of TPW followed by enzymatic hydrolysis. UADA hydrolysis of TPW was optimized by response surface methodology (RSM) at maximum power (900 W) for 2 h. The optimum conditions were determined as 50°C, 1:6 (w/v) solid:liquid ratio, and 1% (w/v) DA concentration, which yielded 20.34 g/L FS concentration. Furthermore, its DA hydrolysis was also optimized by using RSM for comparison and the optimized conditions were found as 120°C, 1:8 solid:liquid ratio, and 1% acid concentration, which produced 25.3 g/L FS yield. Even though the produced sugars with UADA hydrolysis are slightly less, but it can provide significant cost saving due to the lower temperature requirement and less liquid consumption. Besides, enzymatic hydrolysis applied after pretreatments of TPW were very more economic than the conventional enzymatic hydrolysis in the literature due to shorter time requiring. In conclusion, ultrasound‐assisted is a promising technology that can be successfully applied for hydrolysis of biomass and can be an alternative to the other hydrolysis procedures and also TPW can be considered as suitable carbon source for the production of value‐added products like biofuels, organic acids, and polysaccharides.

Yield, quality and biochemical properties of various strawberry cultivars under water stress

BACKGROUND Although strawberry (Fragaria x ananassa Duch.) species are sensitive to abiotic stress conditions, some cultivars are known to be tolerant to different environmental conditions. We examined the response of different strawberry cultivars to water stress conditions in terms of yield, quality and biochemical features. The trial was conducted under two different irrigation regimes: in grow bags containing cocopeat (control, 30%; water stress, 15% drainage) with four different cultivars (Camarosa, Albion, Amiga and Rubygem). RESULTS Fruit weight declined by 59.72% and the yield per unit area by 63.62% under water stress conditions as compared to control. Albion and Rubygem were found to be more tolerant and Amiga the most sensitive in terms of yield under stress conditions. Water stress increased all biochemical features in fruits such as total phenol, total anthocyanin, antioxidant activity and sugar contents. Among the cultivars, glucose and fructose was higher in Albion. CONCLUSION Considering the rise in global warming, identification of resistant and tolerant cultivars to stress conditions are crucial for future breeding programmes. Our results showed that some of the fruits physical features were affected negatively by stress conditions whereas many of the biochemical features such as total anthocyanin content, total phenolic content and antioxidant activity were positively modulated.

Therefore, the present study is undertaken to produce fermentable sugars by using ultrasound-assisted dilute acid hydrolysis of TPW followed by enzymatic hydrolysis. Furthermore, optimization of ultrasound-assisted dilute sulfuric acid (UADA) hydrolysis conditions of TPW was performed by response surface method at maximum power (900 W) for 2 h. Optimized conditions were 50°C, 1:6 (w/v) solid: liquid ratio, and 1% (w/v) dilute acid ratio, which yielded as 20.34 g/L fermentable sugar concentration. In conclusion, ultrasound-assisted dilute acid hydrolysis can be successfully applied for hydrolysis of TPW, which is a favorable carbon source for numerous value-added product productions via microbial fermentation.
