A method is described which makes quantitative studies of the action of Cortisone on connective tissue possible. Foreign body granulomas are provoked in rats by subcutaneous implantation of pellets of compressed cotton. Application of Cortisone results in a diminution of granuloma formation, which can be expressed quantitatively by determining the fresh and the dry weight. Cortisone was effective by local as well as by general application, similar concentrations producing the same degree of inhibition of connective tissue.

Aldosterone applied locally in doses of 50–100 γ enhances slightly the growth of granuloma and in doses of up to 200 γ fails to inhibit such growth. In the doses, then, its action differs from that of hydrocortisone, cortisone, and cortexone.

Experiments have been carried out with the method already described to study the influence of Cortisone on development of foreign body granuloma. The formation of foreign body granuloma and its inhibition by Cortisone are independent of the influence of the hypophysis. It has been found that the granuloma develops in the same way in the hypophysectomized animal as in the normal one. Although in the hypophysectomized animal the inhibition produced by Cortisone is of identical type and degree, it could be concluded that most processes are independent of the influence of the hypophysis.

The action of Triethylenmelamine, an anti-tumour substance, was investigated on tumourgrowth and on development of the foreign body granuloma comparatively. It was found that this substance induces in both cases a similar dosis-dependent inhibition of growth. As the inhibiting action of Triethylenmelamine can even be demonstrated on rapidly growing normal tissues, it is questionable whether the antagonistic action against tumour can be designed as tumour-specific.

The weight has been measured of the granuloma produced after implantation of the same foreign body in the kidneys, the testes, the liver and the subcutaneous tissue of male rats. It was found that granulomae in the testes and liver are nearly twice as heavy as in the kidney or in the subcutaneous tissue. Enhancement or inhibition of this reaction by treating the animals with cortexone shows organ-dependent effects on granuloma formation.

It has been shown that local application of a mixture of precipitating antiserum with homologous plasma or serum promotes markedly the growth of granuloma tissue in the rat.
