ブタ精漿の主な蛋白質は精嚢腺由来である.中国豚の2品種梅山豚と金華豚の精漿蛋白質を超薄層等電点電気泳動法により分析したところ,両者の間に著しい違いが認められた.金華豚では複数のバンドが観察されるのに対して,梅山豚の精漿蛋白質では等電点の高いバンドが主として一本観察された.それらの蛋白質から糖鎖を除去するとSDS-ポリアクリルアミドゲル電気泳動では梅山豚,金華豚共に濃染される二本のバンドとなった.このことから,ブタ精漿蛋白質は糖鎖の異なる2分子の蛋白質より成ることが示された.また,シアル酸の除去により,金華豚型のバンドは梅山豚型となり,糖鎖へのシアル酸の付着数により,品種間の精漿蛋白質の差異が現われるものと考えられた.このことと関連して,梅山豚の精漿蛋白質のシアル酸の含有量は金華豚のそれに比べて低い値を示した.梅山豚と金華豚の交雑F1ブタと,F1ブタと梅山豚との戻し交配ブタの精漿蛋白質の分析の結果,金華豚の示す精漿蛋白質の多様性は梅山豚のそれに対して優性に遺伝すると推定される.

ブタ乳酸脱水素酵素(LDH)の2種類のアイソザイム(LDH-AとLDH-B)のそれぞれのcDNAがブタゲノムDNAの制限酵素断片長多型(RFLPs)検出のための有効なプローブとなり得るか否かを調べた.5種類の制限酵素(Bam HI, Msp I, Pst I, Hae III, Eco RI)で切断したときいずれのプローブを用いても変異が検出された.金華豚と梅山豚の交雑によって得た一家系の親子のそれぞれのDNAをMsp Iで切断して得たRFLPsにっいて分析した結果,LDH-Aで13本,LDH-Bで9本のフラグメントが検出できた.その内,変異を示したのはLDH-Aで9本,LDH-Bで4本であった.また,両親のどちらか一方のみに認められたフラグメントはLDH-Aで6本,LDH-Bで3本,計9本で,その遺伝様相はすべての子豚に遺伝するもの,一部の子豚に遺伝するものとに区分され,前者は親豚でホモの,後者はヘテロ型の状態であったと推定できた.その結果,これら9本のフラグメントは親子鑑定へ利用が可能であった.次に,制限酵素Msp IでブタゲノムDNAを切断し,LDHアイソザイムのcDNAをプローブに用いたRFLPsとM13ファージ反復配列をプローブに用いたDNAフィンガープリント(DFP)の親子鑑定への応用の際の有効性を比較した.DFPでは両親のいずれか一方に存在するフラグメントが7本あり,これらは親子鑑定に利用可能であった.一方,LDHアイソザイムの各cDNAをプローブにして検出されるRFLPsではそれ以上のフラグメントが利用可能で,DFPと比べて親子鑑定の有効性にそん色の無いことが示された.以上の結果から,LDH-AとLDH-Bの両アイソザイムのcDNAをプローブに用いて検出されるRFLPsはブタの個体識別や親子鑑定の際の有効な遺伝的マーカーとなり得ることが判明した.

The paternity test may be necessary even on pasturing beef cattle in order to clarifythe pedigree of offspring, find out a bull carrying undesirable genes resulting in givingbirth to abnormal calves, and study tlte breeding behavior of bulls in pasture.In this investigation, the efficiency of various blood grouping systems for paternitycases involving two or more bulls was estimated from gene frequettcy data in JapaneseShorthorn cattle. The results actually obtained are shown below.(I) The expected probability of making exclusion for paternity cases involving twobulls by nine systems of red-cell antigens was 0.705, artd that by live protein types was0.490. It increased to 0.850, however, when estimated by the combined use of all tlaesystems.(2) In paternity cases involving more than two bulls, the probability of makingexclusion was calculated from the following formula: P.zP.Cn=1>, where P. is the prob-ability of exclusion in two bulls and n is the number of bulls involved. In pasturingJapanese Shorthorn cows with five bulls, the probability of making exclusion decreasedto 0.522.(3) In nineteen actual paternity cases in pasturing cattle, including three possiblebulls, about 84% of the cases were solved by all the systems combined.

Proteinase activity in the sucrose solution extracts of the membrane of ovarian folliele and the liver from hens with atretic ovaries was measured. The atrophy of ovaries was experimentally induced in the normal laying hens by starving them for 4 days. The effects of starvation and vitellin production on the proteinase activity in the liver were also examined with chicks.1. Proteinase activity at pH 4.0 in the extracts of the follicular membrane from the atretic ovaries showed remarkably higher level than that from normal ovaries, but there was no significant difference between the levels of the proteinase activity at pH 8.2 in the extracts of the follicular membrane from normal and atretic ovaries.2. The extracts of the liver from the starved chicks showed higher proteinase activity level at pH 4.0 than that from normal chicks.3. The extracts of the liver from the chicks induced vitellin production by estrogen injection showed lower proteinase activity level at pH 4.0 than that from normal chicks.4. Proteinase activity at pH 4.0 in the extracts of the liver from the starved hens with atretic ovaries showed slightly higher level than that from normal, but the difference was not significant.
