The physiological response of obese rat model with rambutan peel extract treatment

Abstract Objective To determine body weight gain, expression of Igf-1 and Igf-1 receptor on obese rat model treated with rambutan peel extract (RPE) as a physiological response. Methods Normal and obese rat feed with normal and high calorie diet around 12 weeks and continued to treat with ellagic acid, RPE 15, 30 and 60 mg/kg body weight respectively. Physiological responses observed were weight gain and expression of Igf-1 with its receptor. Body weight of rat was weighed once per week. Expression of Igf-1 and igf-1R observed with fluorescence immunohistochemistry. The intensity of Igf-1 and Igf-1R expression was analysis using FSX-BSW software. Results The lowest weight gain was obtained on obese rat model treated with RPE 30 mg/kg body weight. The expression of Igf-1 and Igf-1R were reduced on obese rat model treated with RPE compared with obese rat model of non treatment (P Conclusions The RPE was effecting to the physiological response on obese rat model. The RPE 30 mg/kg body weight inhibited body weight gain and decreased the expression of Igf-1 and Igf-1R of obese rat model.

Rambutan peel extract increases leptin efficiency and decreases lipid peroxidation in a rat model of obesity

Obesity induces oxidative stress and causes disturbances in the production of adipokines. Leptin is a hormone synthesized by adipose cells. Rambutan Peel Extract (RPE) contains polyphenols, which can be used as anti-obesity agents. The objective of this study was to analyze the effect of RPE on leptin levels in visceral fat and liver lipid peroxidation in an obese rat model. Thirty male rats aged 12 weeks were divided into groups: normal rat, obese rat model without treatment, obese rat model with 15 mg/kg RPE (O-RPE15), 30 mg/kg (O-RPE30) and 60 mg/kg (O-RPE60). Leptin efficiency, including leptin levels in visceral fat, was detected indirectly by ELISA and food efficiency was determined. Lipid peroxidation parameters, including malondialdehyde (MDA) levels, were determined by the thiobarbituric acid method (TBA), and PPARγ expression in the liver was detected by immunohistochemistry. Visceral fat leptin levels decreased in the O-RPE30 and O-RPE15 groups. Food consumption and calorie intake were decreased in the O-RPE60, O-RPE15 and O-RPE30 groups, respectively. The greatest food efficiency was observed in the O-RPE15 and O-RPE30 treatments. Liver MDA levels decreased in O-RPE15 and O-RPE30 treatments. There were no differences in PPARγ expression in all the treatments.

Rheumatoid Arthritis (RA) is an autoimmune disease causing chronic inflammatory and degeneration of connective tissue of synovial membrane.1,2 RA disease is usually indicated by swelling on wrist, leg, and knee. Other symptoms of RA patient are fatigue and mild fever.3 The prevalence of RA disease is widespread in the world attacking around 0.5-1% of the population. This disease could attack all ages but commonly at the age of 40-60 years.

POTENSI BUAH MERAH (Pandanus conoideus Lam.) DALAM MENURUNKAN KADAR LDL DARAH TIKUS PUTIH

Penyakit jantung koroner (PJK) terutama disebabkan oleh aterosklerosis karena hiperkolesterolimia. Oksidasi low density lipoprotein (LDL) oleh reactive oxygen species (ROS) sebagai penyebab utama proses aterogenik dapat dicegah dengan kehadiran antioksidan seperti vitamin E. Buah dan sayuran banyak mengandung vitamin. Salah satu buah yang mengandung vitamin E adalah buah merah ( Pandanus conoideus Lam.). Penelitian ini bertujuan untuk mengetahui pengaruh pemberian ekstrak buah merah terhadap kadar LDL darah pada tikus putih ( Rattus norvegicus L.). Jenis penelitian ini adalah penelitian eksperimental laboratorium yang didesain mengikuti Rancangan Acak Lengkap (RAL) dengan 6 perlakuan dan 4 kali ulangan dengan parameter penelitian adalah kadar LDL darah tikus putih. Data dianalisis menggunakan one way anova dilanjutkan dengan uji Duncan 95%. Berdasarkan hasil penelitian, minyak buah merah memiliki kemampuan untuk menurunkan kadar LDL darah tikus putih.
