Possible Antibiofilm Effect of Peptides Derived from IcaR Repressor of Staphylococcus epidermidis Responsible for Hospital-Acquired Sepsis

Liliana Muñoz; Luz Mary Salazar; Stefany Botero; Jeannette Navarrete; Gladys Pinilla

Antimicrobial peptides are alternatives to inhibit biofilm formation, which is mediated by ica ADBC and regulated by IcaR, as a mechanisms of virulence of S. epidermidis involved in hospital-acquired infections. Peptides with biological activity analog to the IcaR protein were designed using the prediction program Antibp; their chemical synthesis was carried out by Nα-Fmoc and the peptides were purified and characterized by RP-HPLC and MALDI-TOF. Red cells were used to determine their hemolytic activity. Peptides named IR1, IR2 and IR3, derived from IcaR, were characterized with a high degree of purity; their hemolytic activity was found to be less than 6 % and they were postulated as candidates with analog activity to the native IcaR repressor against the biofilm formation of S. epidermidis. Their low hemolytic potential, allow them to be use in future in vitro trials for therapeutic use.

Detection of dominants antigens to diagnosis of cisticercosis for immunoelectrotransference (EITB) It is employed lyophilized cyst of Cisticerco cellulosae to prepare antigenic extract of which it is used in the identification of Taenia solliumantigens recognized for IgG antibodies of patients with cisticercosis, for immunoelectrotransfer assay (EITB). It is analyze 193 samples (105 cerebrospinal fluid and 88 sera) from patients with diagnosis of cisticercosis for finding clinic, epidemiologic and inmunienzimatic assay (ELISA). The analysis of the antigenic proteins for electrofiretic, allowed to identify polypeptides of 29, 45, 66, 96 and 116 kDa. The frecuency greates in sera and LCR was 29, 45 and 66 kDa. In conclusion, this immunidominants, will power employ in epidemiologic studies and cisticercosis clinics forWestern blot.

The function of the phagocytic cells is an efficient mechanism for non-specific protection against infectious agents and elimination of dead or senile cells. Cells of the innate immunity, such as neutrophils and macrophages, are responsible for this process. Neutrophils are particularly effective at the initial phases of infections caused by Gram negative and Gram positive bacteria. Macrophages participate in late stages of inflammation phagocyting bacteria and cellular debris. The techniques for evaluation of the phagocytic capacity are laborious, require large blood samples and need
