Abstract ISP-I (myriocin, thermozymocidin) was structurally simplified to give 2-amino-2-alkyl-1,3-propanediols that were also potent immunosuppressants. Among the series, 2-amino-2-pentadecyl-1,3-propanediol most actively prolonged rat skin allograft survival and was more effective than ciclosporin.

2-aminoalchohol immunosuppressants: Structure-activity relationships

Abstract The 2-aminoalcohol series of immunosuppressants showed a bell-shaped relationship between the activity and the carbon number. The ( R )-isomers were more potent than the ( S )-isomers, and ( R )-2-aminohexadecanol displayed comparably activity to a potent immunosuppressant, FTY720.

Abstract 2-Substituted 2-aminoethanol was shown to be the minimum essential structure for the immunosuppressive activity of ISP-i (myriocin, thermozymocidin) by simplification of 2-substituted 2-amino-1,3-propanediol, which was generated by modification of ISP-I. Among the series, 2-amino-4-(4-octylphenyl)butanol hydrochloride displayed comparably potent activity to 2-amino-2-[2-(4-octylphenyl)ethyl]-1,3-propanediol hydrochloride, FTY720.

Antibody against a novel, Myriocin (ISP-I)-based immunosuppressant, FTY720

An antibody was prepared by immunizing rabbits with an ovalbumin conjugate of 2-amino-2-(2-(4-(4-mercaptobutyl)phenyl)ethyl)propane-1,3-diol HCl (AMPD-4), which contains the essential structure of the novel immunosuppressant FTY720. As the antibody reacted to not only AMPD-4, but also FTY720, it should be useful for immunoassay of FTY720 in body fluids, tissues and cells.
